Forurensning av preparater av eukaryote ribosomer renset ved tradisjonelle metoder ved co-rense nukleaser og proteaser påvirker negativt på nedstrøms biokjemiske og strukturelle analyser. En rask og enkel kromatografisk rensing metoden brukes til å løse dette problemet ved hjelp av gjær ribosomer som modell system.
Eukaryote ribosomer er mye mer labil i forhold til deres eubacterial og archael kolleger, og dermed utgjør en betydelig utfordring for forskere. Spesielt problematisk er det faktum at lysis av celler utgivelser et stort antall av proteaser og nukleaser som kan redusere ribosomer. Derfor er det viktig å skille ribosomer fra disse enzymene så raskt som mulig. Dessverre forlater konvensjonelle differensial ultracentrifugation metoder ribosomer utsatt for disse enzymene for uakseptabelt lang tid, påvirker deres strukturelle integritet og funksjonalitet. For å løse dette problemet, benytter vi en kromatografisk metode ved hjelp av en cystein ladet Sulfolink harpiks. Denne enkle og raske søknad reduserer co-rensende proteolytiske og nucleolytic aktiviteter, produsere høy avkastning av intakte, svært biokjemisk aktiv gjær ribosomer. Vi foreslår at denne metoden også bør gjelde for pattedyr ribosomer. Enkelheten avmetoden, og den forbedrede renhet og aktivitet av kromatografisk renset ribosomet representerer en betydelig teknisk fremskritt for studier av eukaryote ribosomer.
