The protocols outlined herein facilitate the convenient investigation of bacterial ethylene responses by utilizing 2-chloroethylphosphonic acid (CEPA). Ethylene is produced in situ through the decomposition of CEPA in an aqueous bacterial growth medium, circumventing the requirement for pure ethylene gas.
Ethylene (C2H4) is a gaseous phytohormone that is involved in numerous aspects of plant development, playing a dominant role in senescence and fruit ripening. Exogenous ethylene applied during early plant development triggers the triple response phenotype; a shorter and thicker hypocotyl with an exaggerated apical hook. Despite the intimate relationship between plants and bacteria, the effect of exogenous ethylene on bacteria has been greatly overlooked. This is partly due to the difficulty of controlling gaseous ethylene within the laboratory without specialized equipment. 2-Chloroethylphosphonic acid (CEPA) is a compound that decomposes into ethylene, chlorine, and phosphate in a 1:1:1:1 molar ratio when dissolved in an aqueous medium of pH 3.5 or greater. Here we describe the use of CEPA to produce in situ ethylene for the investigation of ethylene response in bacteria using the fruit-associated, cellulose-producing bacterium Komagataeibacter xylinus as a model organism. The protocols described herein include both the verification of ethylene production from CEPA via the Arabidopsis thaliana triple response assay and the effects of exogenous ethylene on K. xylinus cellulose production, pellicle properties and colonial morphology. These protocols can be adapted to examine the effect of ethylene on other microbes using appropriate growth media and phenotype analyses. The use of CEPA provides researchers with a simple and efficient alternative to pure ethylene gas for the routine determination of bacterial ethylene response.
Het olefine etheen (C 2 H 4) werd ontdekt als een plantenhormoon in 1901 toen het werd vastgesteld dat erwt zaailingen, gekweekt in een laboratorium steenkolengas lampen gebruikt, vertoonde een abnormale morfologie waarbij stengels (hypocotylen) waren korter, dikker en boog zich zijwaarts ten opzichte van de normale erwt zaailingen; een fenotype later aangeduid als de triple respons 1,2. Latere studies toonden aan dat ethyleen een belangrijke plantenhormoon dat talrijke ontwikkelingsprocessen zoals groei, stressrespons, rijping van fruit en senescence 3. Arabidopsis thaliana, modelorganisme voor plantenbiologie onderzoek regelt, werd bestudeerd met betrekking tot de reactie op ethyleen. Verschillende ethyleen reactie mutanten zijn geïsoleerd door het benutten van de triple respons fenotype waargenomen in het donker-volwassen A. thaliana zaailingen in aanwezigheid van etheen 1,4,5. De biosynthetische voorloper voor ethyleen productie in planten is 1-aminocyclopropane carbonzuur (ACC) 6 en wordt vaak gebruikt in de drievoudige respons assay endogene etheenproduktie die leidt naar de drievoudige respons fenotype 1,4,5 verhogen.
Hoewel de ethyleenreactie schaal bestudeerd in planten, is het effect van exogeen ethyleen op bacteriën zeer weinig bestudeerd ondanks de nauwe associatie van bacteriën met planten. Een studie meldde dat bepaalde Pseudomonas stammen kunnen overleven met behulp van ethyleen als enige bron van koolstof en energie 7. Echter slechts twee studies aangetoond dat de bacteriën reageren op ethyleen. De eerste studie toonde aan dat stammen van Pseudomonas aeruginosa, P. fluorescens, P. putida en P. syringae waren chemotactische richting van ethyleen met gebruik van agarose plug assay waarin gesmolten agarose werd gemengd met een chemotaxis buffer geëquilibreerd met zuiver ethyleengas 8. Echter, voor zover ons bekend, zijn er geen furth geweester rapporten met behulp van zuiver ethyleengas om bacteriële ethyleen reactie, waarschijnlijk te karakteriseren als gevolg van de moeilijkheid van de behandeling van gassen in het laboratorium zonder gespecialiseerde apparatuur. Het tweede rapport van bacteriële ethyleen reactie aangetoond dat ethyleen verhoogde bacteriële cellulose productie en beïnvloed genexpressie in de fruit-geassocieerde bacterie, Komagataeibacter (voorheen Gluconacetobacter) xylinus 9. In dit geval, de ethyleen vrijmakende verbinding, 2-chloroethylphosphonic acid (CEPA) werd gebruikt om ethyleen in situ te produceren binnen het bacteriële groeimedium, het omzeilen van de behoefte aan zuivere ethyleengas of gespecialiseerde apparatuur.
CEPA ethyleengas in een 1: 1 molaire verhouding boven pH 3,5 10,11 via een base gekatalyseerde eerste orde reactie 12-14. De afbraak van CEPA is positief gecorreleerd met pH en temperatuur 13,14 en resulteert in de productie van ethylalcoholeen, chloride en fosfaat. CEPA biedt onderzoekers die geïnteresseerd zijn in het bestuderen van bacteriële reacties op ethyleen met een handig alternatief voor gasvormige ethyleen.
Het algemene doel van de volgende protocollen wordt een eenvoudige en efficiënte werkwijze voor bacteriële ethyleenreactie bestuderen en inclusief validering van fysiologisch relevante niveaus etheenproductiefaciliteiten van CEPA decompositie in bacteriële groeimedium analyse cultuur pH te waarborgen CEPA ontleding, niet verminderd tijdens de groei van bacteriën, en de evaluatie van het effect van ethyleen op de bacteriële morfologie en fenotype. We laten deze protocollen het gebruik van K. xylinus echter deze protocollen kunnen worden aangepast ethyleenrespons bij andere bacteriën bestuderen door het geschikte groeimedium en fenotype analyse.
De hier beschreven werkwijzen beschrijven de in situ bereiding van ethyleen uit CEPA voor de studie van bacteriële ethyleenreactie met het modelorganisme, K. xylinus. Deze werkwijze is zeer nuttig als ethyleen kan worden verkregen door aanvulling elk waterig medium dat een pH heeft hoger dan 3,5 10,11 met CEPA ontkennen de noodzaak van zuivere ethyleengas of gespecialiseerde laboratoriumapparatuur.
