Hier beschrijven we massaspectrometrieanalyse om de alomtegenwoordigheid van EYFP-CENP-A K124R mutant te identificeren, wat suggereert dat alomtegenwoordigheid bij een andere lysine wordt veroorzaakt door de EYFP-tagging in het CENP-A K124R gemuteerd eiwit. Lysine 306 (K306) alomtegenwoordigheid in EYFP-CENP-A K124R werd met succes geïdentificeerd, wat overeenkomt met lysine 56 (K56) in CENP-A door middel van massaspectrometrie analyse. Een voorbehoud wordt besproken in het gebruik van GFP /EYFP of het taggen van eiwit met een hoog moleculair gewicht als hulpmiddel om de functie van een eiwit te analyseren. De methode van massaspectrometrie analyse hier gepresenteerd kan worden toegepast op menselijke CENP-A eiwit met verschillende tags en andere centromere-kinetochore eiwitten. Deze combinatorische methoden bestaande uit verschillende tests / analyses kunnen worden aanbevolen voor onderzoekers die geïnteresseerd zijn in het identificeren van functionele rollen van alomtegenwoordigheid.
In de meeste eukaryoten moeten spindelmicrotubuli zich hechten aan één enkel gebied van elk chromosoom, centromere genoemd. De kinetochore is een complex van eiwitten die zich op de centromere bevinden. Het bestuderen van de timing van centromere en kinetochore eiwit bewegingen en de structuur van kinetochores en centromeres is belangrijk voor het begrijpen van chromosoom instabiliteit (CIN) en kanker progressie. De belangrijkste vragen zijn hoe de chromosomale locatie en functie van een centromere (d.w.z. centromere identiteit) worden bepaald en hoe ze deelnemen aan nauwkeurige chromosoomsegregatie. In de meeste soorten, de aanwezigheid van een speciaal nucleosoom met een specifieke histone-achtige eiwit genaamd CENP-A definieert de centromere identiteit. Daarom wordt voorgesteld dat CENP-A de niet-DNA-indicator (epigenetisch merk) van centromere identiteit is. Het is belangrijk om het mechanisme op te helderen van hoe CENP-A de centromere identiteit bij mensen definieert.
De Holliday junction recognition protein (HJURP) is de CENP-A-specifieke chaperon die CENP-A in centromerische nucleosomen1,2,3deponeert . We have previously reported that the CUL4A-RBX1-COPS8 E3 ligase is required for CENP-A ubiquitylation on lysine 124 (K124) and centromere localization4. Ook onze resultaten toonden aan dat de centromere werving van nieuw gesynthetiseerde CENP-A vereist reeds bestaande ubiquitylated CENP-A5. Thus, a model was provided suggesting that CENP-A ubiquitylation is inherited through dimerization between cell divisions.
In tegenstelling tot onze bevindingen en die van Yu et al., werden de negatieve resultaten betreffende CENP-A en zijn centromeric lokalisatie onlangs gepubliceerd6. Het artikel beweerde dat CENP-A wijzigingen op lysine 124 (K124) zijn onmisbaar voor de oprichting, onderhoud, en de functie op lange termijn van de menselijke centromeres, op basis van hun negatieve resultaten waaruit blijkt dat de mutatie van K124R niet van invloed CENP-A centromere lokalisatie noch cel levensvatbaarheid6. Er is echter voldoende ruimte voor discussie in hun resultaten en conclusies, en we hebben al beschreven welk probleem er zou kunnen zijn in hun vorige publicatie7. Er moet op worden gelet dat ze eiwitten hebben versmolten met CENP-A, die veel grotere moleculaire gewichten hebben dan de grootte van endogene CENP-A: bijvoorbeeld, ze gesmolten ~ 30 kDa verbeterde gele fluorescente eiwitten (EYFP) tot ~ 16 kDa CENP-A en analyseerde EYFP-CENP-A K124R fusie eiwit in hun RPE-1 CENP-A-/F knock-out systeem. K124 ubiquitine zal naar verwachting niet rechtstreeks binden aan HJURP op basis van structurele voorspellingen4,maar de toevoeging van mono-ubiquitine zal naar verwachting een impact hebben op de eiwitcontenschap van CENP-A. Het eiwit van CENP-A-conformatie kan worden veranderd door de aanwezigheid van een groot fusie-eiwit, en deze conformatieverandering kan de structurele veranderingen maskeren die worden veroorzaakt door het verlies van alomtegenwoordigheid. Wij stellen voor dat de fusie van groot-en klein-grote eiwit alome alomtegenwoordigheid induceert bij een andere lysine dan K124 in EYFP-CENP-A K124R mutant en deze alomtegenwoordigheid op een andere plaats remt / maskeert de oorspronkelijke K124R single mutant fenotype. Bewijs dat alomtegenwoordigheid optreedt bij verschillende lysine in de CENP-A K124R mutant eiwit met een large tag eiwit (EYFP) werd gemeld in onze vorige publicatie8. Er werd vastgesteld dat EYFP tagging alomtegenwoordigheid induceert van een andere lysinesite van EYFP-CENP-A K124R en dat EYFP-CENP-A K124R mutant bindt aan HJURP. Als gevolg hiervan remt/maskeert deze alomtegenwoordigheid op een andere locatie het oorspronkelijke K124R enkel mutantene fenotype, en zowel EYFP-CENP-A WT als K124R mutanten vertoonden centromere lokalisatie (we gebruikten en vergeleken pBabe-EYFP-CENP-A WT en K124R mutant, samen met pBabe-EYFP control.). The results demonstrated that Flag-tagged or untagged CENP-A K124R mutants are lethal but can be rescued by a monoubiquitin fusion, suggesting that CENP-A ubiquitylation is indispensable to cell viability.
In de afgelopen jaren hebben veel studies verschillende tests ontwikkeld om posttranslationele modificaties (PTMs) van CENP-A-eiwit en andere centromere-kinetochore-eiwitten te identificeren, zowel in vivo als in vitro9,10,11. Analoog aan de PTMs van histoneiwitten die een belangrijk mechanisme zijn dat de functie van chromatine regelt, zijn PTMs van centromerische chromatinecomponenten ook betrokken bij een essentieel mechanisme om de algehele structuur en functie van centromeres te reguleren. De meeste CENP-A PTM-sites zijn specifiek voor CENP-A-bevattende nucleosomen, hoewel een paar daarvan in histone H3 worden bewaard, wat suggereert dat wijziging van deze residuen bijdraagt aan de centromere-specifieke functie. PTMs van CENP-A met inbegrip van fosforylatie, acetylatie, methylatie, en alomtegenwoordigheid werden eerder gemeld9, wat suggereert dat CENP-A wordt onderworpen aan een verscheidenheid van PTM’s en hun combinatorische arrays op zijn amino terminus en C-terminus histone-fold domein. Het belang van CENP-A wijzigingen in meerdere functies werd onthuld door vele groepen, waaronder de onze. Deze functies omvatten CENP-A depositie bij centromeres, eiwitstabiliteit en rekrutering van het CCAN (constitutief centromere-geassocieerd netwerk)9. Beperkte studies en bevindingen van CENP-A PTM’s zijn echter voorgevormd wanneer vergelijkingen worden gemaakt met een van canonieke histonen die hun functie direct of indirect reguleren. Technische rapporten die zich richten op de methodologie om deze CENP-A PTMs te identificeren zijn ook beperkt.
