We bieden een gedetailleerd protocol voor een alomtegenwoordyleringstest van een specifiek substraat en een E3-ubiquitine-ligase in zoogdiercellen. HEK293T-cellijnen werden gebruikt voor eiwitoverexpressie, het polyubiquityleerde substraat werd gezuiverd uit cellysaten door immunoprecipitatie en opgelost in SDS-PAGE. Immunoblotting werd gebruikt om deze posttranslationele modificatie te visualiseren.
