Bu protokol, Epstein-Barr virüs parçacıklarının, forbol 12-miristat 13-asetat ile viral litik döngüyü indükledikten sonra insan P3HR1 hücre hattından izole edilmesine izin verir. DNA daha sonra viral preparattan çıkarılır ve viral parçacık konsantrasyonunu ölçmek için gerçek zamanlı PCR'ye tabi tutulur.
