University of Tulsa 3 articles published in JoVE Environment Simulation of Early Earth Hydrothermal Chimneys in a Thermal Gradient Environment Ninos Hermis1, Gabriel LeBlanc2, Laura M. Barge1 1NASA Jet Propulsion Laboratory, California Institute of Technology, 2The University of Tulsa The goal of this protocol is to form simulated hydrothermal chimneys via chemical garden injection experiments and introduce a thermal gradient across the inorganic precipitate membrane, using a 3D printable condenser that can be reproduced for educational purposes. Genetics CRISPR/Cas9 Editing of the C. elegans rbm-3.2 Gene using the dpy-10 Co-CRISPR Screening Marker and Assembled Ribonucleoprotein Complexes. Amy Smith1, Mary Bergwell*1, Ellie Smith*1, Danita Mathew*1, Jyoti Iyer1 1Department of Chemistry and Biochemistry, The University of Tulsa The goal of this protocol is to enable seamless CRISPR/Cas9 editing of the C. elegans genome using assembled ribonucleoprotein complexes and the dpy-10 co-CRISPR marker for screening. This protocol can be used to make a variety of genetic modifications in C. elegans including insertions, deletions, gene replacements and codon substitutions. Immunology and Infection Combining Analysis of DNA in a Crude Virion Extraction with the Analysis of RNA from Infected Leaves to Discover New Virus Genomes Jeanmarie Verchot1, Aastha Thapa2, Dulanjani Wijayasekara3, Peter R. Hoyt4 1Texas A&M Agrilife Center at Dallas, 2Noble Research Center, Oklahoma State University, 3Department of Biology, College of Engineering and Natural Sciences, The University of Tulsa, 4Bioinformatics and Genomics Core Facility, Department of Biochemistry and Molecular Biology, Oklahoma State University Here we present a new approach to identify plant viruses with double-strand DNA genomes. We use standard methods to extract DNA and RNA from infected leaves and carry out next-generation sequencing. Bioinformatic tools assemble sequences into contigs, identify contigs representing virus genomes and assign genomes to taxonomic groups.
