Hansen1, Simon Hippenmeyer1 1Institute of Science and Technology Austria A protocol to perform lineage tracing and functional genetic analysis of candidate genes at a single cell level using mosaic analysis with double markers (MADM) is presented. MADM clonal analysis provides a quantitative framework to measure the proliferative behavior, cellular output, and lineage relationship of individual progenitors and their daughter cells. Neuroscience A Micro-CT-based Method for Characterizing Lesions and Locating Electrodes in Small Animal Brains Javier Masis1,2, David Mankus2, Steffen B.E. Cox1,2 1Department of Molecular and Cellular Biology, Harvard University, 2Center for Brain Science, Harvard University, 3Department of Organismic and Evolutionary Biology, Harvard University, 4Institute of Science and Technology Austria This article describes a straightforward method to prepare small animal brains for micro-CT imaging, in which lesions can be quantified and electrodes located with high precision in the context of the whole brain. Biology Light Sheet Fluorescence Microscopy of Plant Roots Growing on the Surface of a Gel Daniel von Wangenheim1, Robert Hauschild2, Jiří Friml1 1Developmental and Cell Biology of Plants, Institute of Science and Technology Austria, 2Bioimaging Facility, Institute of Science and Technology Austria This protocol shows a plant sample preparation method for light-sheet microscopy. The setup is characterized by mounting the plant vertically on the surface of a gel and letting it grow in controlled bright conditions. This allows long-term observation of plant organ development in standardized conditions.
