Das Video zeigt die orthotopische Inokulationstechnik von Brustkrebs in einem Mausmodell. Diese Technik erzeugt eine hohe Tumorigeneserate mit weniger Variabilität in Tumorgröße und -form im Vergleich zu subkutaner oder nicht-chirurgischer Injektion.
Protocol
1. Präparation von Zellen (für 10 Mäuse) HINWEIS: Die Zellen sollten innerhalb von 1 h geimpft werden, nachdem sie von der Schale gelöst wurden, um eine verminderte Zelllebensfähigkeit zu vermeiden. Insbesondere sollte die Zellsuspension innerhalb von 15 min nach dem Ablösen der Zellen von der Schale in die gelatineartige Proteinmischung gemischt werden, um ihre Lebensfähigkeit zu erhalten.
