The vascular endothelial cells play a significant role in many important cardiovascular disorders. This article describes a simple method to isolate and expand endothelial cells from the mouse aorta without using any special equipment. Our protocol provides an effective means of identifying mechanisms in endothelial cell physiopathology.
The vascular endothelium is essential to normal vascular homeostasis. Its dysfunction participates in various cardiovascular disorders. The mouse is an important model for cardiovascular disease research. This study demonstrates a simple method to isolate and culture endothelial cells from the mouse aorta without any special equipment. To isolate endothelial cells, the thoracic aorta is quickly removed from the mouse body, and the attached adipose tissue and connective tissue are removed from the aorta. The aorta is cut into 1 mm rings. Each aortic ring is opened and seeded onto a growth factor reduced matrix with the endothelium facing down. The segments are cultured in endothelial cell growth medium for about 4 days. The endothelial sprouting starts as early as day 2. The segments are then removed and the cells are cultured continually until they reach confluence. The endothelial cells are harvested using neutral proteinase and cultured in endothelial cell growth medium for another two passages before being used for experiments. Immunofluorescence staining indicated that after the second passage the majority of cells were double positive for Dil-ac-LDL uptake, Lectin binding, and CD31 staining, the typical characteristics of endothelial cells. It is suggested that cells at the second to third passages are suitable for in vitro and in vivo experiments to study the endothelial biology. Our protocol provides an effective means of identifying specific cellular and molecular mechanisms in endothelial cell physiopathology.
Das vaskuläre Endothel nicht nur eine Barriereschicht ist , die Blut und Gewebe trennt, wird eine große endokrine Drüse betrachtet , die mit einer Oberfläche von 400 Quadratmetern 1 über den gesamten Gefäßbaum erstreckt. Das Wohlbefinden des Endothels ist wesentlich für vaskuläre Homöostase. Die dysfunktionalen Endothels beteiligt sich an verschiedenen kardiovaskulären Erkrankungen wie Atherosklerose, Vaskulitis und Ischämie / Reperfusion Verletzungen, usw. 2-4. Bis heute sind die spezifischen zellulären und molekularen Mechanismen in dieser Krankheitseinstellungen beteiligt sind nicht gut verstanden durch die diffuses anatomischen Natur des Endothels.
Die Maus ist ein wichtiges Modell für die Forschung, da genetische Manipulationstechniken mehr bei Mäusen entwickelt als in jeder anderen Säugetierspezies. Jedoch ist die Isolierung von primären Maus-Aorta-Endothelzellen als besonders schwierig, da die geringe Größe der Aorta enzymat machtic Verdauung von Endothel nicht praktikabel. Einige Verfahren berichtet , zu isolieren und zu reinigen ECs 5-7 erfordern.
Das Ziel dieses Protokolls ist es, ein einfaches Verfahren zu verwenden, um Endothelzellen aus der Maus Aorta zu isolieren und zu erweitern, ohne eine spezielle Ausrüstung. In diesem Protokoll wird die frisch isolierten Aorta in kleine Segmente geschnitten und ausgesät auf einer Matrix mit dem Endothel für endothelial Austrieb zu ermöglichen nach unten zeigt. Nach Segmenten entfernt werden, werden Endothelzellen in Endothel-bevorzugtes Medium erweitert und sind für Experimente nach zwei oder drei Passagen bereit. Die Vorteile des beschriebenen Verfahrens sind, dass: 1) beträchtlich hohe Anzahl von Endothelzellen aus einer einzigen Aorta geerntet werden; 2) die Lebensfähigkeit der Zellen ist gut erhalten; und 3) keine spezielle Ausrüstung oder Technik erforderlich ist. Sie stellt ein wirksames Mittel zur spezifischen zellulären und molekularen Mechanismen in Endothelzellen Pathophysiologie identifizieren. Für diejenigen, die bei der Untersuchung pr interessiert sindimary kultivierten Endothelzellen aus entweder Gen-Knock-out-Mäuse, Gen-Knock-in-Mäusen, oder einem murinen Krankheitsmodell ist dieses Protokoll sehr nützlich und leicht zu praktizieren.
Diese Studie zeigt, ein einfaches Verfahren zur Isolierung und Kultur Endothelzellen von einer Maus Aorta ohne spezielle Ausrüstung. Die Immunfluoreszenzfärbung zeigte, daß die Mehrzahl der Zellen Endothelzellen nach dem zweiten Durchgang waren. Es wird vorgeschlagen , dass die Zellen am zweiten in den dritten Durchgang geeignet sind , in vitro und in vivo Experimenten endothelial biology zu studieren.
