A potential general method for the synthesis of water-soluble multimetallic peptidic arrays containing a predetermined sequence of metal centers is presented.
We demonstrate a method for the synthesis of a water-soluble multimetallic peptidic array containing a predetermined sequence of metal centers such as Ru(II), Pt(II), and Rh(III). The compound, named as a water-soluble metal-organic complex array (WSMOCA), is obtained through 1) the conventional solution-chemistry-based preparation of the corresponding metal complex monomers having a 9-fluorenylmethyloxycarbonyl (Fmoc)-protected amino acid moiety and 2) their sequential coupling together with other water-soluble organic building units on the surface-functionalized polymeric resin by following the procedures originally developed for the solid-phase synthesis of polypeptides, with proper modifications. Traces of reactions determined by mass spectrometric analysis at the representative coupling steps in stage 2 confirm the selective construction of a predetermined sequence of metal centers along with the peptide backbone. The WSMOCA cleaved from the resin at the end of stage 2 has a certain level of solubility in aqueous media dependent on the pH value and/or salt content, which is useful for the purification of the compound.
Gecontroleerde synthese van complexe moleculaire structuren is altijd al een groot probleem in de synthetische chemie geweest. Vanuit dit oogpunt, op meerkernige heterometallische complexen synthetiseren ontwerpbaar wijze nog een waardige onderwerp in het gebied van anorganische chemie vanwege het aantal mogelijke structurele resultaten van de ligand-metallatie gebaseerde benadering die gewoonlijk wordt gebruikt kan worden betwist de bereiding van monomere metaalcomplexen. Alhoewel verschillende voorbeelden van meerkernige heterometallische complexen dusver 1,2,3 gemeld, de trial-and-error te verbieden, hun synthese vereist de ontwikkeling van een eenvoudige methode die toepasbaar voor uiteenlopende structuren.
Als een nieuwe benadering van dit probleem aan te pakken, in 2011 hebben we melding gemaakt van een synthetische methodologie 4,5 waarbij verschillende mononucleaire metaalcomplexen met een Fmoc-beschermd aminozuur groep opeenvolgend zijn gekoppeld met meerdere op te gevenmetallic peptidische arrays met de protocollen van vaste-fase-polypeptidesynthese 6. Vanwege de aard van opeenvolgende polypeptide synthese een specifieke sequentie van meerdere metaalcentra is rationeel designable door het regelen van het aantal en de volgorde van de koppelingsreacties van monomeren metaalcomplexkleurstoffen. Later, deze benadering werd verder modulair diverse grotere en / of vertakte structuren matrix te combineren met de covalente binding tussen twee kortere arrays 7.
Hier zullen we laten zien hoe de synthese van dergelijke multimetallic peptide arrays doorgaans wordt beheerd door het kiezen van de onlangs gemeld WSMOCA (1 8 CAS RN 1827663-18-2; figuur 1) als een representatief voorbeeld. Hoewel de synthese van een bepaalde reeks wordt in dit protocol, dezelfde procedures worden voor de synthese van een groot aantal verschillende sequenties, waaronder isomeren 9. We verwachten dat deze protocol zal onderzoekers inspiratie te nemen aan de wetenschap van sequentie gecontroleerde verbindingen, waarbij de moleculen onderzocht tot nu toe gewoonlijk zijn biopolymeren maar zelden omvatten voorbeelden van metaal-complex-gebaseerde soorten.
Perfecte verwijdering van de ongewenste chemische stoffen van de hars is niet altijd mogelijk door eenvoudig wassen met oplosmiddelen die gemakkelijk deze stoffen kan oplossen. Een belangrijke techniek om de hars doeltreffend te wassen moet veroorzaken om te zwellen en krimpen herhaaldelijk zodat de chemicaliën die nog in worden gedwongen. Daarom heeft de hars in onze procedure wordt met CH 2 Cl 2 en MeOH beurtelings als het wordt gewassen (bijvoorbeeld protocol 2.1.4).
