sistemi optogenetic utilizzano proteine geneticamente codificate che cambiano conformazione in risposta a specifiche lunghezze d'onda della luce per alterare i processi cellulari. Vi è la necessità per la coltura e misurare sistemi che incorporano programmati illuminazione e la stimolazione dei sistemi optogenetic. Questo protocollo può essere implementato in molti laboratori familiarità con crescente coltura cellulare e microscopia, e l'apparecchiatura utilizzata è poco costoso e fatto di componenti prontamente disponibili. Un recipiente di coltura trasparente è posizionato sopra una matrice di diodi emettitori di luce (LED) in grado di emettere 1 W / cm 2 -10 mW / cm 2 di luce. I microbi sono coltivate nel recipiente di coltura in continuo; una pompa peristaltica viene utilizzata per aggiungere media alladiluizione, un'altra viene utilizzata per prelevare cultura ad un tasso minore al microscopio, e la differenza sfugge attraverso un'uscita di overflow. Una piastra elettrica mantiene la temperatura. L'aria viene continuamente pompata nel recipiente di coltura per mantenere una pressione positiva nonché per mescolare ed aerare la cultura. Per dimostrare l'applicazione di questo protocollo, abbiamo misurato la risposta alla luce diverse intensità di cellule di Saccharomyces cerevisiae ingegnerizzate con un respon luce bluve sistema optogenetic che controlla la trascrizione della proteina fluorescente. S. cerevisiae, comunemente noto come lievito di birra, è stato scelto perché più sistemi optogenetic per controllare l'espressione genica in questo sistema esistono già 14, 15, 16.
