Here, we present a protocol for hydraulic extrusion of the spinal cord as well as identification and isolation of specific dorsal root ganglia (DRGs) in the same rodent. Compared to standard spinal cord isolation methods, this method is significantly faster and reduces the risk of tissue damage.
Traditionally, the spinal cord is isolated by laminectomy, i.e. by breaking open the spinal vertebrae one at a time. This is both time consuming and may result in damage to the spinal cord caused by the dissection process. Here, we show how the spinal cord can be extruded using hydraulic pressure. Handling time is significantly reduced to only a few minutes, likely decreasing protein damage. The low risk of damage to the spinal cord tissue improves subsequent immunohistochemical analysis. By performing hydraulic spinal cord extrusion instead of traditional laminectomy, the rodents can further be used for DRG isolation, thereby lowering the number of animals and allowing analysis across tissues from the same rodent. We demonstrate a consistent method to identify and isolate the DRGs according to their localization relative to the costae. It is, however, important to adjust this method to the particular animal used, as the number of spinal cord segments, both thoracic and lumbar, may vary according to animal type and strain. In addition, we illustrate further processing examples of the isolated tissues.
Het algemene doel van deze methode is het ruggenmerg isoleren alsook DRG 1 identificeren en te isoleren, 2. Hydraulische extrusie van het ruggenmerg is een beduidend snellere methode dan de traditionele manier ruggenmerg isolatie door laminectomie, dwz door het breken van de wervelkolom een voor een, en dit reduceert de kans op weefselbeschadiging veroorzaakt door de dissectie proces 3, 4 . DRG kan moeilijk te identificeren zijn. Correcte identificatie is van groot belang voor tissue-analyse, bijvoorbeeld na heupzenuw letsel. Nummering van de DRG volgens hun lokalisatie ten opzichte van de costae, kan DRG consistent geïdentificeerd.
Weefsel geïsoleerd door technieken die hier getoond kunnen worden toegepast op een groot aantal analyses zoals Western blotting 5,6, 7 en qPCR immunohistochemische kleuring 8.
Bij de identificatie DRG, is het belangrijk om rekening te houden dat bekend het aantal ruggemerg segmenten te variëren met diersoort en stam 9, 10, 11. De voordelen bij het uitvoeren van deze werkwijze in muizen het grote aantal genetisch gemodificeerde varianten beschikbaar en de relatief lage kosten behuizing. De voordelen in het gebruik van ratten zijn de relatief hoge opbrengst en weefsel als betrekken zenuw verwondingen, zal de procedure worden verlicht met de grootte.
Wanneer de wervelkolom wordt verstoord, bijvoorbeeld door cervicale dislocatie, de ruggenmerg splitsen tijdens extrusie. Als het ruggenmerg kan worden geëxtrudeerd, kan de wervelkolom iets afgesneden buiten en binnen de extrusie poging kan worden herhaald. Indien het gehele ruggemerg noodzakelijk voor verdere analyse, namelijk bestaande uit de cervicale uitbreiding en de lumbale vergroting, de wervelkolom moet alleen worden bijgesneden. Als alleen de lumbale vergroting noodzakelijk voor verdere analyse, moet het ruggenmerg worden getrimd volgens de onderhavige protocol. De wervelkolom moet worden rechtgetrokken met behulp van de vingertoppen aan de extrusie te verlichten.
Het protocol geldt voor knaagdieren van alle leeftijden en wordt hier geïllustreerd door een volwassene muizen (8 weken), een muis pup (5 dagen) en volwassen ratten (10 weken). Afhankelijk van het type dier en de stam, kan het aantal thoracale en lumbale secties variëren 9, </sup> 10, 11. Afhankelijk van de eiwitten te analyseren, volwassen knaagdieren worden geperfuseerd met enzym remmende oplossingen voorafgaand aan euthanasie. Als het uitvoeren van een experiment met eenzijdige zenuwletsel, kan het ruggenmerg worden gesplitst in ipsilaterale en contralaterale zijden met behulp van ultra-fijne pincet direct na de extrusie. Verder kan elke zijde worden gesplitst in dorsale en ventrale zijden. Het weefsel kan vervolgens worden verwerkt voor verdere analyses, zoals Western blotting.
Transcardial perfusie-fixatie met PFA vóór ruggenmerg extrusie moet worden vermeden, aangezien dit maakt het ruggenmerg niet flexibel en voorkomt ruggenmerg hydraulische extrusie. Hydraulische ruggenmerg extrusie zal afscheuren de dorsale wortels. Voor experimenten waarbij bevestigd dorsale wortels nodig zijn, wordt laminectomie aanbevolen. Bovendien worden spinale hersenvliezen verloren hydraulische extrusie, die kan worden vermeden door standaard laminectomie <sup class = "xref"> 3.
Hydraulische extrusie van het ruggenmerg en DRG isolatie kan ook worden uitgevoerd met geoxygeneerd kunstmatige cerebrospinale vloeistof (ACSF) plaats ijskoude PBS. Het gebruik van ACSF maakt behoud van het geïsoleerde weefsel in een betere fysiologische omgeving, hetgeen vooral van belang bij latere elektrofysiologische opnames 12, 13. Een alternatief kan zijn ACSF PBS dat 1 g / l glucose voor het genereren van primaire DRG culturen 14.
Hydraulische extrusie van het ruggenmerg is een beduidend snellere methode dan de traditionele manier ruggenmerg isolatie door laminectomie, waardoor weefsel doorlooptijd en dus minder kans op schade aan eiwitten. Perfusie met een hechtmiddel voorafgaand aan het isoleren van het ruggenmerg door laminectomie kan het risico van weefselbeschadiging tijdens dissectie en tijdens de laatste verwijdering van het verminderenruggenmerg van de wervelkolom. Echter, weefsel fixatie de weg staat aan de toepasselijkheid ervan op analyses, zoals Western blotting. Hydraulische extrusie opbrengsten structureel beschadigd weefsel 3 geschikt voor een breder scala van analyses.
Consistente identificatie van DRG's kan moeilijk zijn. Dit is echter essentieel voor weefselanalyse, bijvoorbeeld na heupzenuw letsel. Nummering van de DRG volgens hun lokalisatie ten opzichte van de costae, kan DRG consistent geïdentificeerd. Kleuring van ruggenmergweefsel en DRG kunnen worden geoptimaliseerd door middel van de geïllustreerde weefselbehandeling variaties in Protocol stap 7.
