I detta protokoll presenterar vi en experimentell design med hjälp av en villkorlig knockdown system och en anpassad sfär bilda analys att studera effekten av clusterin på stemness av patient-härledda GCSCs. Protokollet kan enkelt anpassas för att studera både in vitro- och in vivo-funktion hos stemness-associerade gener i olika typer av CSC.
