Denne protokollen beskriver metoden, materialer, utstyr og trinn for nedenfra og opp forberedelse av RNA og proteinproduserende syntetiske celler. Det indre akdige rommet i de syntetiske cellene inneholdt S30 bakteriell lysat innkapslet i en lipid bilayer (dvs. stabile liposomer), ved hjelp av en vann-i-olje emulsjon overføringsmetode.
