Nya strategier för att troget modellera somatiska mutationer i hematopoetiska stam- och stamceller (HSPC) är nödvändiga för att bättre studera hematopoetisk stamcellsbiologi och hematologiska maligniteter. Här beskrivs ett protokoll för att modellera heterozygota gain-of-function-mutationer i HSPCs genom att kombinera användningen av CRISPR/Cas9 och dubbel rAAV-givartransduktion.
