Adam Mickiewicz University 2 articles published in JoVE Biology Combining Clearing and Fluorescence Microscopy for Visualising Changes in Gene Expression and Physiological Responses to Plasmodiophora brassicae Deeksha Singh1, Sara Blicharz1, Karolina Stefanowicz1, Laura Ragni2, Kornel Michalak3, Agnieszka Bagniewska-Zadworna3, Robert Malinowski1 1Integrative Plant Biology Team, Institute of Plant Genetics Polish Academy of Sciences, 2ZMBP-Center for Plant Molecular Biology, University of Tübingen, 3Department of General Botany, Institute of Experimental Biology, Faculty of Biology, Adam Mickiewicz University The present protocol describes an optimized method for the histological observation of galls induced by Plasmodiophora brassicae. Vibratome sections of hypocotyls are cleared before fluorescence imaging to study the involvement of transcription factors and phytohormones during disease progression. This protocol overcomes resin embedding limitations, enabling in planta visualization of fluorescent proteins. Chemistry Surface Properties of Synthesized Nanoporous Carbon and Silica Matrices Angelina Sterczyńska1, Małgorzata Śliwińska-Bartkowiak1,2, Małgorzata Zienkiewicz-Strzałka3, Anna Deryło-Marczewska3 1Faculty of Physics, Adam Mickiewicz University, 2NanoBioMedical Centre, 3Department of Physicochemistry of Solid Surface, Faculty of Chemistry, Maria Curie-Skłodowska University Here we report the synthesis and characterization of ordered nanoporous carbon (with a 4.6 nm pore size) and SBA-15 (with a 5.3 nm pore size). The work describes the surface and textural properties of nanoporous molecular sieves, their wettability, and the melting behavior of D2O confined in the materials.
