Vi beskriver procedurer for mærkning og genotypebestemmelse nyfødte mus og generere primære neuronale kulturer fra dem. Genotypebestemmelse er hurtig, effektiv og pålidelig og giver mulighed for automatiseret nukleinsyre-ekstraktion. Dette er især nyttigt for neonatally dødbringende mus og deres kulturer, der kræver forudgående afslutning af genotypning.
Høj opløsning analyse af morfologi og funktion af pattedyr neuroner kræver ofte genotypebestemmelse af de enkelte dyr, efterfulgt af en analyse af primære kulturer af neuroner. Vi beskriver en række procedurer for: mærkning nyfødte mus, der skal genotypebestemmes, hurtig genotypning, og oprettelse af lav densitet kulturer af hjernens neuroner fra disse mus. Individuelle mus er mærket med tatovering, der giver mulighed for langvarig identifikation varig ind i voksenalderen. Genotypebestemmelse af den beskrevne protokol er hurtig og effektiv, og giver mulighed for automatiseret ekstraktion af nukleinsyre med god pålidelighed. Dette er nyttigt under omstændigheder, hvor tilstrækkelig tid til konventionel genotypning ikke er tilgængelig, f.eks, i mus, der lider af neonatal dødelighed. Primære neuronale kulturer genereres ved lav densitet, hvilket muliggør billeddannelse eksperimenter ved høj rumlig opløsning. Denne protokol kan anvendes til mus med andre genetiske mutationer, såvel som andre dyr. Desuden kan de enkelte komponenter i protokol bruges til isolerede delprojekter. Således denne protokol vil have brede programmer, ikke kun i neurovidenskab, men også på andre områder af biologiske og medicinske videnskaber.
Gnavermodeller for genetiske sygdomme har vist sig nyttige for fysiologiske funktioner normale proteiner og nukleinsyrer samt de patofysiologiske konsekvenser af defekter i disse. Eksempler indbefatter mus deficiente for proteiner involveret i vigtige cellulære funktioner, samt musemodeller for lidelser, såsom Alzheimers sygdom. Imidlertid kan visse genetiske manipulationer føre til neonatal letalitet kort eller nogle få dage efter fødslen. I disse tilfælde primære cellekulturer er et vigtigt redskab, fordi levende celler kan opnås fra de embryonale eller neonatale hvalpe før døden, kan de opretholdes i mindst et par uger in vitro, og i denne periode tidlige neuronal udvikling kan følges af biokemiske, funktionelle og morfologiske forsøg. For de primære kulturer, kan det være gavnligt at pladen neuroner ved lav densitet; dette gør det muligt at visualisere den enkelte somata, dendritter, axonale aksler og nerve terminaler med høj rumlig opløsning. Imidlertid kræver typisk overlevelse og differentiering af neuroner ved lav densitet, at de er belagt på en glial fødelag, co-dyrket med gliaceller i fravær af fysisk kontakt med dem eller dyrket i medium konditioneret med glia 1.
Etableringen af low-density neuronale kulturer på gliale fødelag kan være afhængig af hurtig og pålidelig genotypebestemmelse forhånd – inden for et par timer i modsætning til et par dage. Hastighed er især vigtigt, når den neuronale genotype skal modsvares med den for en glial fødelag forberedt på forhånd. Som et mere praktisk eksempel kan det være nødvendigt at bestemme, hvilke unger af hvilken genotype til anvendelse ved generering kulturer, for at optimere effektiviteten af et eksperiment.
Her demonstrere vi arbejder protokol, der er blevet brugt til hurtig, forenklet og pålidelig mus genotypning i tidligere udgivelser 2-6. Mus haler oget kommercielt tilgængeligt kit anvendes. Denne protokol omfatter enkelt-trins ekstraktion af nukleinsyrer fra vævet, og som hverken kræver en nukleinsyre-oprensningstrin eller anvendelse af en afslutning buffer ("stop opløsning"). Pålideligheden af denne genotypebestemmelse metode illustreres ved at præsentere resultaterne af en række tests, hvor forskellene bliver introduceret i forhold til grundbeløbet for prøverne, at dyrenes alder og længden af PCR-amplikonerne. Dette sæt giver fordelene ved automatiseret ekstraktion og pålidelighed.
Af hensyn til at være omfattende, er anvendelsen af tatovering langsigtet identifikation af genotypede mus også påvist. Tatovering opnås ved at anvende tatovering blæk til dermis i huden (under epidermis) 7. En fremgangsmåde er beskrevet til tatovering af trædepuder på nyfødte eller 1 dag gamle mus, selv om tatoveringer kan anvendes på andre dele af kroppen, såsom haler og tæer, og Animals i alle aldre. Desuden vil procedurer påvises for udpladning og dyrkning mus neuroner ved en lav massefylde, baseret på optimeret fremstilling af forskellige typer af glia fødelag 2,8.
Vi bruger en genetisk musemodel af den nedarvede neurologiske lidelse DYT1 dystoni – en autosomal dominerende bevægelse lidelse forårsaget af en mutation i genet TOR1A (c.904_906delGAG / c.907_909delGAG; p.Glu302del / p.Glu303del) 9. Det kodede protein, Torsina, tilhører de "ATPaser forbundet med diverse cellulære aktiviteter" (AAA +) familie af proteiner, hvis medlemmer normalt udfører chaperone-lignende funktioner, som bistår i: proteinudfoldning, protein-kompleks demontering, membran handel og vesikelfusion 10-13. De mutationen resulterer i en i-ramme deletion af en codon for glutaminsyre, og kan føre til manifestation af 'tidlig debut generaliseret isoleret dystoni' 14,15. Men stienophysiological mekanismer, der er ansvarlige for denne lidelse er fortsat dårligt forstået. I en knock-in musemodel, mutantallelen er Tor1a tm2Wtd, nævnt i det følgende som Tor1a AE. Heterozygot AE-Torsina knock-i mus er levedygtige og genetisk efterligne menneskelige patienter med DYT1 dystoni, hvorimod homozygot knock-i mus dør efter fødslen 16,17, med latenstiden til postnatal død påvirket af genetisk baggrund 18. Den tidlige død homozygot knock-i mus kræver, at både genotypebestemmelse af dyr og etablering af neuronale kulturer afsluttes hurtigt. Som et andet eksempel på genotypebestemmelse Tfap2a (transkriptionsfaktor AP-2α, aktiverende enhancer protein 2α) vil blive anvendt. Proteinet kodet af dette gen er vigtig i reguleringen af flere cellulære processer, såsom proliferation, differentiering, overlevelse og apoptose 19.
Protokollen præsenteres her indeholder procedurer til tatovering at mærke / identificere mus, for genotype mus fra hale tips, og for dyrkning mus hjernen neuroner ved lav tæthed. I en runde af forsøg med 6-8 unger, disse procedurer kræver typisk ~ 0,5 timer, ~ 4 timer og ~ 2 timer, henholdsvis på i alt 6-7 timer. Dette gør det praktisk for en enkelt eksperimentatoren at udfylde alle de nødvendige procedurer fra tidspunktet for hvalpenes fødsel til plettering af neuronale kulturer – på mindre end en enkelt arbe…
