Synthesis of human monoclonal antibodies is the first step in studies aimed at unraveling the pathophysiological mechanisms of auto-antibody-mediated immune responses. We have developed a protocol to generate recombinant human immunoglobulin G (IgG) monoclonal antibodies from blood sorted B cells, including B-cell isolation, antibody cloning and in vitro synthesis.
Finding new methods for generating human monoclonal antibodies is an active research field that is important for both basic and applied sciences, including the development of immunotherapeutics. However, the techniques to identify and produce such antibodies tend to be arduous and sometimes the heavy and light chain pair of the antibodies are dissociated. Here, we describe a relatively simple, straightforward protocol to produce human recombinant monoclonal antibodies from human peripheral blood mononuclear cells using immortalization with Epstein-Barr Virus (EBV) and Toll-like receptor 9 activation. With an adequate staining, B cells producing antibodies can be isolated for subsequent immortalization and clonal expansion. The antibody transcripts produced by the immortalized B cell clones can be amplified by PCR, sequenced as corresponding heavy and light chain pairs and cloned into immunoglobulin expression vectors. The antibodies obtained with this technique can be powerful tools to study relevant human immune responses, including autoimmunity, and create the basis for new therapeutics.
Målet med denne artikkelen er å beskrive i detalj en metode for å generere og karakter humane IgG monoklonale antistoffer hentet fra humane perifere mononukleære blodceller (PBMC).
Interessen for å studere humane antistoffer har vokst i mange ulike forskningsfelt. Spesielt mange forskningsgrupper er interessert i patologi forårsaket av auto-antistoffer 1-3. Vi har klonet og karakterisert patogene autoantistoffer 1. Studiet av auto-antistoffer kan bidra til å identifisere sine mål og utvikle terapeutiske strategier, for eksempel ved å bruke konkurren antistoffer 4. Videre kan studiet av humane antistoffer også være av interesse i andre dvs. innen forskning, for å vurdere immunresponsen etter vaksinering 5, for å karakterisere antistoffprofil av individer som ble eksponert og ble motstandsdyktig mot visse patogener 6, eller for å studere hvilke antistoffer er idet naturlige repertoaret 7,12.
Flere teknikker er blitt utviklet for å generere rekombinante humane monoklonale antistoffer 8-12; De fleste av disse bruke fag-display og B-celle immortalisering. Bruken av fagdisplay har blitt mye brukt for oppdagelsen av nye antistoffer 13. Men den har en stor ulempe, nemlig at de tunge og lette kjede parene av det humane immunoglobulin blir dissosiert i prosessen. Fremstilling av hybridomer med humane B-celler eller EBV-transformasjon overvinner denne ulempe.
Vi bruker infeksjon av tymiske B-celler med EBV i kombinasjon med polyklonale B-celle stimulering via Toll-like receptor 9 (TLR-9) 6,12.
I denne artikkelen beskriver vi i detalj den teknologien som vi bruker for utvikling av IgG humane antistoffer, med en komplett oversikt over alle trinnene fra PBMC isolasjon til in vitro antistoff generasjon. DetteProtokollen kan brukes for analyse av hvilken som helst type av humant IgG-profil. I vårt laboratorium, er B-celler som produserer IgG-antistoffer blitt separert fra resten av PBMC etter sortering. Femti sortert B-celler 8 kan deretter bli sådd ut i multi-brønnplater og udødeliggjort ved EBV og TLR-9-aktivering, for klonal ekspansjon av enkelt B-celler.
