A quick protocol for proteolytic digestion with an in-house built flow-through tryptic microreactor coupled to an electrospray ionization (ESI) mass spectrometer is presented. The fabrication of the microreactor, the experimental setup and the data acquisition process are described.

Identifiseringen og karakteriseringen av rensede proteiner blir ofte oppnådd ved hjelp av MS-teknikker. Proteinet blir spaltet med et enzym og dets peptider blir ytterligere analysert ved MS ved hjelp av en enkel tilførsel eksperimentelt oppsett. Massespektrometer kan utføre analyse og bekreftet sekvensen til et protein innenfor en kort tidsramme. Med moderne datainnhentingsmetoder, kan denne prosessen gjøres wTVen på noen minutter eller sekunder. Den begrensende faktor i å fullføre hele prosessen på en kort tidsskala er den proteolytiske spaltning trinnet. Til tross for de mange fordelene med immobilisert enzymreaktorer, vil kostnadene ved kommersielle patroner er høy, er tilgjengeligheten av microfluidic enheter for rutinemessig bruk begrenset, og bruk av mikrobølgeovn eller PCT teknologier medfører behov for ekstra instrumentering. Målet med dette arbeidet var å utvikle en metode som circumvents disse ulemper, og som enkelt kan implementeres i alle laboratorier for å styrke forskere med en enkel og effektiv metode for å utføre enzymatisk spaltning av proteiner i forberedelse for MS-analyse i løpet av minutter. Tilnærmingen er avhengig av bruk av hydrofobe, C18-partikler som er forhåndsinstallert i en kapillær eller mikrofluid enhet, og adsorpsjonen av protein (er) av interesse på disse partiklene, fulgt av enzymatisk nedbrytning under infusjonen av enzymet over hele pakket seng og fanges protein (er).
