This manuscript describes an ex vivo model system comprised of organ-conditioned media derived from the lymph node, bone, lung, and brain of mice. This model system can be used to identify and study organ-derived soluble factors and their effects on the organ tropism and metastatic behavior of cancer cells.
Breast cancer preferentially metastasizes to the lymph node, bone, lung, brain and liver in breast cancer patients. Previous research efforts have focused on identifying factors inherent to breast cancer cells that are responsible for this observed metastatic pattern (termed organ tropism), however much less is known about factors present within specific organs that contribute to this process. This is in part because of a lack of in vitro model systems that accurately recapitulate the organ microenvironment. To address this, an ex vivo model system has been established that allows for the study of soluble factors present within different organ microenvironments. This model consists of generating conditioned media from organs (lymph node, bone, lung, and brain) isolated from normal athymic nude mice. The model system has been validated by demonstrating that different breast cancer cell lines display cell-line specific and organ-specific malignant behavior in response to organ-conditioned media that corresponds to their in vivo metastatic potential. This model system can be used to identify and evaluate specific organ-derived soluble factors that may play a role in the metastatic behavior of breast and other types of cancer cells, including influences on growth, migration, stem-like behavior, and gene expression, as well as the identification of potential new therapeutic targets for cancer. This is the first ex vivo model system that can be used to study organ-specific metastatic behavior in detail and evaluate the role of specific organ-derived soluble factors in driving the process of cancer metastasis.
Bröstcancer är den vanligaste diagnostiserade cancerformen hos kvinnor och den näst vanligaste orsaken till cancerrelaterade dödsfall 1. Bröstcancer höga dödligheten är främst på grund av fel i konventionell behandling för att minska och eliminera metastatisk sjukdom; ungefär 90% av cancerrelaterade dödsfall beror på metastaser 2. Att förstå de underliggande molekylära mekanismerna för metastaserande kaskad är avgörande för utvecklingen av läkemedel effektiva i både tidigt och sent skede bröstcancer.
Tidigare forskning har bidragit till att belysa den flerstegs karaktär av bröstcancer metastaser och det antas att resultatet av både cancer progression och metastasering är till stor del beroende på samspelet mellan cancerceller och värdmiljön 3. Kliniska observationer tyder på att många cancerformer visar organ tropism, dvs. Tendensen att företrädesvis metastasera till specifika organs.In CASe av bröstcancer, sprider en patients sjukdom typiskt eller sprider sig till 5 viktigaste platserna, inklusive ben, lungor, lymfkörtlar, lever och hjärna 4-6. Många teorier har utvecklats för att förklara denna process, men bara ett fåtal har stått emot tidens tand. Ewings teori om metastas, som föreslås i 1920-talet, hypotesen thatthe distribution av metastaser var strikt på grund av mekaniska faktorer; varigenom tumörceller sker i hela kroppen av normala definierade fysiologiska blodflödesmönster och helt enkelt stoppa i första kapillär sängen de möter 7. I motsats, Stephen Pagets 1889 "utsäde och jord" hypotes föreslog att ytterligare molekylära interaktioner var ansvariga för överlevnad och tillväxt av metastaser, varigenom cancerceller ( "frön") endast kan etablera sig och proliferatein organmikromiljöer som producerar lämpliga molekylära faktorer ( "jord ") 8. Nästan ett sekel senare, Leonard Weiss enligttog en meta-analys av tidigare publicerade obduktions uppgifter och bekräftade Ewings förutsägelse som många metastaserande tumörer som upptäcks vid obduktion hittades i de förväntade proportioner som skulle förväntas om metastatisk organ tropism bestämdes genom blodflödesmönster ensam. Men i manyinstances det fanns färre eller fler metastaser bildas vid vissa platser då kan förväntas av Ewings föreslagna mekaniska faktorer 9. Dessa konton och teorier tyder på att specifika mikromiljöer organ spelar en avgörande roll i spridningsmönster och efterföljande tillväxt och överlevnad av många cancerformer, bland annat bröstcancer.
Tidigare forskning har främst fokuserat på tumörcells härledda faktorer och deras bidrag till organ tropism observerats vid bröstcancer metastaser 10-12, men lite forskning har undersökt faktorer som härrör från orgelmikro som kan ge en gynnsam nisch för inrättandetav bröstcancermetastaser. Detta är till stor del tillskrivas de tekniska utmaningarna att studera komponenterna i orgelmikro in vitro.
Den aktuella artikeln beskrivs en omfattande ex vivo modellsystem för att studera påverkan av lösliga komponenter i lymfkörtel, ben, lunga och hjärna på den metastatiska beteendet hos humana bröstcancerceller. Tidigare studier har validerat detta modellsystem genom att visa att olika bröstcancercellinjer visar cellinje specifik och organspecifika malignt beteende som svar på organkonditionerade media som motsvarar deras in vivo metastatisk potential 13. Detta modellsystem kan användas för att identifiera och utvärdera specifika organhärledda lösliga faktorer som kan spela en roll i den metastatiska beteendet hos bröst- och andra typer av cancerceller, inklusive påverkan på tillväxt, migration, stjälkliknande beteende och genuttryck, samt identifiering avpotentiella nya terapeutiska mål för cancer. Detta är den första ex vivo modellsystem som kan användas för att studera organspecifik metastatisk beteende i detalj och för att utvärdera rollen av organhärledda lösliga faktorer i att driva processen för cancermetastas.
Metastas är en komplex process genom vilken en serie av cellulära händelser är ytterst ansvarig för vävnadsinvasion och avlägsna tumören etablering 4,30,31.
