振動や音波などの非局在的な機械的力は、発生から恒常性まで、さまざまな生物学的プロセスに影響を与えます。動物は行動を変えることによってこれらの刺激に対処します。このような行動修正の根底にあるメカニズムを理解するには、関心のある行動中の神経活動の定量化が必要です。ここでは、特定の周波数、変位、および持続時間の非局在振動を有する カエノラブディティス・エレガンス を自由に行動させる際のカルシウムイメージングの方法を報告する。この方法は、音響トランスデューサを使用して十分に制御された非局在振動を生成し、単一細胞分解能で誘発されたカルシウム応答を定量化することを可能にする。原理の証明として、振動に対する C.エレガンスの 脱出応答の間、単一の介在ニューロンAVAのカルシウム応答が実証される。このシステムは、機械的刺激に対する行動応答の根底にある神経メカニズムの理解を容易にする。
動物はしばしば、振動や音響波などの非局在的な機械的刺激にさらされる1,2。これらの刺激は恒常性、発達、生殖に影響を与えるため、動物はそれらに対処するために行動を変えなければなりません3,4,5。しかし、そのような行動改変の根底にある神経回路およびメカニズムは、ほとんど理解されていない。
線虫のメカノセンティック行動、Caenorhabditis elegansは単純な行動パラダイムであり、ワームは通常、非局在的な振動に遭遇すると、前方の動きから後方の脱出応答に行動を変える6。この挙動の根底にある神経回路は、主に5つの感覚ニューロン、4対の介在ニューロン、および数種類の運動ニューロン7、8から構成される。さらに、ワームは、反復刺激9、10、11を含む間隔をあけた訓練の後、そのような機械的刺激に慣れる。したがって、この単純な行動応答は、非局在的な振動誘発行動と記憶の両方の根底にある神経メカニズムを調査するための理想的なシステムを構成する。非局在振動の影響下で自由に行動するワームにおけるカルシウムイメージングのためのプロトコルが示されている。以前に報告されたシステムと比較して、このシステムは追跡のために追加のカメラを必要としないという点で簡単です。ただし、非局在振動の周波数、変位、および持続時間を変更することができます。AVA介在ニューロンの活性化は後方脱出応答を誘導するため、AVA特異的プロモーターの制御下でカルシウム指示薬GCaMPとカルシウム非感受性蛍光タンパク質TagRFPを共発現するワームを例に挙げた(詳細は材料表参照)。このプロトコルは、ワームが前方から後方への移動に切り替える際のAVAニューロンの活性化を示しています。このプロトコルは、メカノ感覚行動の根底にある神経回路メカニズムの理解を容易にする。
