Vi præsenterer her, en protokol til at lade nøjagtig kvantificering af mitokondriel DNA (mtDNA) methylering. I denne protokol beskriver vi en Enzymatisk nedbrydning af DNA med BamHI kombineret med en bioinformatic analyse pipeline, som kan bruges til at undgå overvurdering af mtDNA methylering niveauer forårsaget af mtDNA sekundær struktur.
