Cet article présente une série de méthodes consécutives pour l’expression et la purification de Salmonella typhimurium tryptophan synthase comp ce protocole un système rapide pour purifier le complexe protéique en une journée. Les méthodes couvertes sont la mutagénèse dirigée par site, l’expression des protéines chez Escherichia coli,la chromatographie d’affinité, la chromatographie par filtration sur gel et la cristallisation.
