Questo protocollo descrive un minigene reporter assay per monitorare l'impatto delle mutazioni del sito di giunzione 5'-splicing sullo splicing e sviluppa snRNA U1 soppressore per il salvataggio dell'inibizione dello splicing indotta da mutazioni. I costrutti di snRNA U1 reporter e soppressore sono espressi in cellule HeLa e lo splicing viene analizzato mediante estensione di primer o RT-PCR.
