Este protocolo descreve um ensaio de repórter minigene para monitorar o impacto de mutações de 5'-splices no splicing e desenvolve snRNA supressor U1 para o resgate da inibição de emenda induzida por mutação. As construções de snRNA do repórter e supressor U1 são expressas em células HeLa, e o splicing é analisado por extensão de primer ou RT-PCR.
