细胞骨架是相互作用的生物聚合物的动态复合网络,为细胞提供结构和机械支持。相关的分子马达和结合蛋白重组和调整细胞骨架,使细胞生长、改变形状、变硬、移动甚至自我修复,从而实现从迁移和分裂到机械传感的无数细胞过程1,2。除了在细胞生物物理学中的重要性之外,细胞骨架也是活性物质的典型例子,其潜在材料应用范围从伤口愈合和药物输送到过滤和软机器人1,3,4,5,6,7,8,9。
赋予细胞骨架独特的结构和机械多样性以及多功能性的两个关键特征是:1)其复合性质,包括多种相互作用的蛋白质丝,例如半柔性肌动蛋白丝和刚性微管,以及它们相关的结合和交联蛋白3,5,10;2)它能够通过肌球蛋白和驱动蛋白等耗能电机连续重组、移动、粗化和执行工作,推动和拉动丝状蛋白1,7,11,12,13。虽然这种优雅的复杂性使细胞骨架能够介导细胞运动、细胞分裂和伤口愈合等多种过程3,6,7,11,但它阻碍了研究人员在体外重建系统中重现细胞骨架体内特征的能力。
目前的前沿重建工作集中在缠结和交联的肌动蛋白丝和微管3,10,14,15,16,17,产生力的肌动蛋白网络2,8,18,19,20,21的复合物,以及由驱动蛋白 – 微管驱动的活性向列相互作用22,23,24,25,26。稳态肌动蛋白-微管复合材料已被证明具有紧急机械性能15,16,27,例如与单组分系统相比,灯丝流动性增强和刚度增加27。对体外肌动肌蛋白系统的研究报告了广泛的结构和动力学特性,这些特性取决于肌动蛋白、肌球蛋白和交联剂的浓度28,29,30,31。例如,在充分交联的情况下,肌动球蛋白网络经历大规模收缩和粗化2,28,30,32,33,34,35,36,而没有交联剂,网络表现出快速,不稳定的流动和破裂19,29.据报道,使用驱动蛋白马达簇交联和拉动微管束的基于微管的重组主动向列具有持久的湍流、延伸、屈曲、破裂和愈合12,22,23,24,25,37,38,39,40,41, 42,43,44,45,46,47.
最近,与无交联剂的肌动球蛋白网络表现出17,26,48的无序流动和网络破裂相比,由肌球蛋白II微丝驱动的肌动蛋白-微管复合材料已被证明会导致更有序的收缩和网络完整性。此外,当肌动蛋白和微管以相当的浓度存在时,复合材料稳健性和力生成的组合得到优化。配方空间这一区域的关键新兴特征包括增强的机械强度26,肌动蛋白和微管的协调运动26,稳定的持续收缩和中尺度重组17。
在这里,描述了设计和调整微管和肌动蛋白丝的共纠缠和交联复合材料,这些微管和肌动蛋白丝分别被肌球蛋白II迷你丝和驱动蛋白簇推出平衡,分别作用于肌动蛋白丝和微管(图1)。这类复合材料的动力学、结构和力学可以通过灯丝、电机和交联剂的相对浓度进行调整,以表现出丰富的平流和湍流、各向同性收缩、加速、减速、解混、硬化、松弛和破裂的相空间。这项工作的重点是制备和调整这类活性细胞骨架复合材料。然而,为了帮助研究人员对所描述的活性复合材料进行基准测试和表征,还详细介绍了使用多光谱共聚焦显微镜的有效成像方法。最后,介绍了可用于测量复合材料动力学、结构和力学的关键计算分析方法的结果。鼓励研究人员采用这些方法,包括差分动态显微镜(DDM),空间图像自相关(SIA)和粒子图像测速法(PIV),因为它们已经过优化,可以表征复合材料的复杂动力学和结构多样性17,26,49。
下面描述的步骤侧重于制备复合材料并使用共聚焦显微镜对其进行成像。描述采集后数据分析和光镊测量的协议可以在以前的著作17,26,48,50中找到,并可根据要求提供。所有材料均列在提供的材料表中。
上述重组系统的一个关键进步是其模块化和可调性,因此鼓励用户修改蛋白质、马达、交联剂等的浓度以适应他们想要的结果,无论是模拟特定的细胞过程还是设计具有特定功能或机械性能的材料。肌动蛋白和微管蛋白浓度范围的限制由聚合肌动蛋白(~0.2μM)57,58,59和微管蛋白(~3 – 4μM)60所需的临界浓度设定在下限,上限由肌动蛋白丝(~90μM)61,62或微管(~35μM)63向向列排列的过渡设定。.肌动蛋白单体和微管蛋白二聚体应聚合成细丝,而不是聚合后混合在一起,以确保它们形成均匀互穿的渗透网络,相互协同支持。复合材料表现出的新动力学依赖于这种相互作用。虽然遵循协议中概述的所有步骤以成功重现显示的结果通常很重要,但有些步骤更严格,而其他步骤则有修改和调整的空间以适应特定需求和可用资源。
例如,确保结果可重现的一个重要步骤是按照材料表中提供的指南正确制备和储存试剂。细胞骨架蛋白(肌动蛋白,微管蛋白,肌球蛋白,驱动蛋白)不稳定,应等分,用液氮快速冷冻,并以一次性等分试样储存在-80°C。从-80°C取出后,应将等分试样保存在冰上。细胞骨架蛋白在额外的冻融循环后不能可靠地保持功能。
微管比肌动蛋白对解聚和变性更敏感。一旦从-80°C中取出,微管蛋白应在聚合前保持在冰上,并在12小时内使用。一旦聚合,微管应保持在室温下。用紫杉醇稳定微管以防止解聚也很重要。肌动蛋白丝的鬼笔环肽稳定对于抑制与肌球蛋白和驱动蛋白活性竞争的ATP消耗肌动蛋白跑步机同样重要。
肌球蛋白马达的超速离心是另一个关键步骤,因为它可以去除不活跃的肌球蛋白死头。不去除酶无活性的单体会导致肌动蛋白网络的被动交联和活性损失。为了延长马达的ATP酶活性,可以结合ATP再生系统,如磷酸肌酸和肌酸磷酸激酶64 。
最后,保持复合活性需要抑制灯丝和电机吸附到样品室壁上,这可以通过显微镜盖玻片和载玻片的钝化来实现。运动蛋白特别容易吸附,这导致复合材料被拉到样品室表面,移出视场,塌陷到2D,不再进行活动。对盖玻片和载玻片进行硅烷化是钝化表面并防止吸附的有效方法(见步骤1)。在体外细胞骨架实验中有效使用的另一种钝化方法是在表面涂覆脂质双层,类似于细胞膜18。如果希望将蛋白质束缚在表面或引入其他特定的蛋白质 – 表面相互作用,则这种方法是有利的,因为双层可以功能化。对于光学镊子实验,微球的钝化也很关键,可以通过碳化二亚胺交联剂化学48用BSA或PEG包覆羧化微球来实现。
研究人员可以考虑更改所提出的协议的几个方面以满足他们的需求。首先,研究人员可以选择用生物交联剂代替非天然生物素-NA交联剂,例如分别交联肌动蛋白和微管的α-肌动蛋白或MAP6528,65,66。与天然交联剂相比,在本文描述的复合材料中使用非天然交联剂的动机是其增强的重现性、稳定性和可调性。由于强大的生物素-NA键,交联剂可以被认为是永久性的,而不是大多数以广泛的周转率瞬时结合的天然交联剂。瞬态交联的动力学使解析交联剂和电机对动力学的贡献变得复杂。此外,生物素-NA接头可用于交联肌动蛋白和微管,以及将肌动蛋白交联到微管。通过这种方式,可以在交联基序之间进行明确的比较,保持所有其他变量(例如,交联剂大小,结合亲和力,化学计量等)不变。最后,掺入生物素-NA接头所需的试剂已广泛商业化,表征良好,并且通常用于许多生物物理实验室。然而,这里描述的体外平台的关键优势之一是其模块化,因此研究人员应该能够选择用天然接头无缝替换生物素-NA接头。
其次,在当前的方案中,肌动蛋白单体和微管蛋白二聚体在加入样品室之前在离心管中聚合成细丝。将缠绕的丝状蛋白溶液流入样品室可能会导致流动对齐,尤其是微管的流动对齐,从而破坏复合材料所需的各向同性和均匀性。事实上,先前关于稳态肌动蛋白-微管复合材料的工作的一个重大进展是能够原位(在样品室中)共聚合肌动蛋白和微管,以确保形成肌动蛋白和微管的各向同性互穿网络15,16,27。然而,将这种方法扩展到活性复合材料需要在肌动蛋白和微管蛋白聚合之前将马达添加到样品中,并在实验前将整个样品在37°C下孵育在一起。对方案的这种变异的测试导致肌动蛋白聚合减少,并且没有明显的运动活性,这可能是由于竞争性ATP酶活性和马达的37°C长时间孵育。幸运的是,遵循当前协议时,复合材料没有明显的流动对齐,如图2、图3和图6所示。尽管如此,鼓励研究人员设计允许原位形成活性复合材料的方案。
另一个需要考虑的点是荧光标记方案,它需要稀疏地标记网络中的所有肌动蛋白丝和微管。这种标记方法经过优化,可以直接可视化网络的结构,而不是通过示踪丝或微球 推断 结构和动力学。然而,权衡是单个细丝没有明亮的标记和可解析。研究人员可以采取的一种方法来解析单丝以及可视化网络结构,方法是在用另一种荧光团标记的预成型细丝中掺杂,这样周围的网络和单个细丝都可以同时成像。然而,当使用两个以上的荧光基团和激发/发射通道时,通道之间的渗漏通常很难消除,因此在选择荧光团、滤光片和激光强度时必须小心。
一个相关的限制是无法可视化复合材料中的肌球蛋白或驱动蛋白马达。使用的荧光标记肌动蛋白单体和微管蛋白二聚体是市售的,而复合材料中肌球蛋白或驱动蛋白的可视化需要内部标记。鼓励研究人员采取下一步来标记电机,就像之前所做的那样18,67,以便能够明确地将电机活动和结合与我们的复合材料所表现出的动力学和结构联系起来。
最后,重要的是要注意,在当前的方案中,驱动蛋白活性的开始和持续时间不受控制。如上所述,由于肌球蛋白活性是使用布比他汀的光失活来控制的,以建立类似的驱动蛋白光激活,因此可以加入光激活ATP。
为了增加这里描述的设计的复杂性,更好地模拟细胞条件并拓宽动态结构功能参数空间,未来的工作将集中在结合中间细丝,如vimentin68,69,以及其他电机,如动力蛋白13,70。凝胶索林也将以不同的浓度掺入以控制肌动蛋白长度14,以及tau蛋白以控制微管硬度。
