未知の情報が豊富であるだけでなく、3次元(3D)の視点から洗練された細胞小器官の微細構造の動的特徴を理解することは、機構研究にとって重要です。電子顕微鏡(EM)は、優れたイメージング深度を提供し、高解像度の画像スタックの再構築を可能にして、ナノメートルスケールでも細胞小器官の微細構造形態を調査できます。したがって、3D再構築は、その比類のない利点のために重要性を増しています。走査型電子顕微鏡(SEM)は、連続したスライスの同じ関心領域から大きな構造を3Dで再構築できるハイスループット画像取得技術を提供します。したがって、細胞小器官の真の3D微細構造を復元するための大規模な3D再構成におけるSEMの適用はますます一般的になっています。このプロトコルでは、膵臓癌細胞のミトコンドリアクリステを研究するために、連続超薄切片と3D再構成技術の組み合わせを提案します。これらの技術がどのように実行されるかの詳細は、オスミウム-チオカルボヒドラジド-オスミウム(OTO)法、シリアル超薄切片イメージング、および視覚化表示を含む、このプロトコルで段階的に説明されています。
ミトコンドリアは細胞内で最も重要な細胞小器官の1つです。それらは細胞の生体エネルギーと代謝の中心的なハブとして機能し1,2、癌において重要な役割を果たします3。膵臓がん(PC)は、その急速な広がりと高い死亡率のために、治療が最も困難ながんの1つです4。主にミトコンドリアの形態の変化によって引き起こされるミトコンドリア機能障害3,5,6,7は、PC8の根底にある疾患メカニズムに関連しています。ミトコンドリアも非常に動的であり、これはネットワーク接続とクリステ構造の頻繁かつ動的な変化に反映されています9。クリステ構造の再形成は、ミトコンドリア機能と細胞状態に直接影響を与える可能性があり10,11、腫瘍細胞の成長、転移、および腫瘍微小環境の変化中に著しく変化します12,13。
近年、科学者たちはEM観察14,15,16,17を用いてこの細胞小器官を研究しています。たとえば、研究者は3D再構成技術を使用してミトコンドリアのダイナミクスを分析しました6、7、18、19。電子顕微鏡画像の3D再構成の一般的な概念と方法は、早くも196820に正式に確立され、電子顕微鏡、電子回折、およびコンピューター画像処理を組み合わせてT4ファージテールを再構築することが含まれていました。これまで、電子顕微鏡3Dイメージング技術は、画像解像度21、自動化度22、処理量23の面で大きな進歩を遂げ、組織レベルからナノメートルスケールのオルガネラ微細構造レベル24まで、生物学研究においてますます幅広い規模で使用されてきました。近年、電子顕微鏡3Dイメージングも幅広い用途の有望な技術となっています25、26、27。
ミトコンドリアクリステへの注目の高まりは、特に微細構造体積イメージングの本質的な要件を示しています。透過型電子顕微鏡(TEM)は、銅グリッド(400メッシュ)28に集められたサンプルを視覚化するために使用され、電子ビームはセクションを通過します。しかしながら、銅グリッドの範囲が限られているため、同じサンプル29の連続スライスを完全に画像化することは不可能である。これは、TEMイメージング中の標的構造の研究を複雑にする。さらに、TEMは、複数のスライスの切断と収集、およびそれらを順次イメージングするなど、時間がかかり、エラーが発生しやすい手動タスクに依存しているため21、大量のサンプルの微細構造再構成には適していません23。現在、TEMカメラアレイ(TEMCA)30 や2台の第2世代TEMCAシステム(TEMCA2)31など、短時間でハイスループットなイメージングを自動化する専用装置を使用することで、大容量サンプルイメージングの高解像度再構成を実現しています。ただし、このタイプのイメージングには、カスタマイズされた機器が必要なため、入手が容易で普遍的であるという利点はありません。
TEMと比較して、SEM32、33に基づいて大面積の数千のシリアル体積画像を自動的に生成する方法は、シリアルイメージングの効率および信頼性を高め、より高いz解像度を提供する34。たとえば、シリアルブロック面走査電子顕微鏡(SBF-SEM)と集束イオンビーム走査電子顕微鏡(FIB-SEM)はどちらも、高速、効率、解像度で微細構造の3D再構成を実現することを可能にしました35,36。ただし、SBF-SEMのダイヤモンドナイフまたはFIB-SEM33,37の集束イオンビームによるフライス加工によって、ブロック表面が機械的に削り取られることは避けられません。試料に対する2つの方法の破壊性のために、さらなる分析のために同じ標的構造を再び再構築することは不可能である38、39、40。さらに、EMを使用して癌細胞の3Dオルガネラ微細構造を再構築し、病理学的変化を観察することを試みた研究はほとんどありません12。このような理由から、膵臓がん細胞などのがん細胞の病態メカニズムをさらに解明するために、ウルトラミクロトームと電界放出型走査電子顕微鏡(FE-SEM)を用いて、ミトコンドリアの微細構造をクリステレベルで解析する新しい断面像を3次元再構成する技術を提案する。この技術により、効率的でアクセスしやすい方法で高解像度のデータを取得できます。ウルトラミクロトームを使用して作られた連続超薄切片は、グリッドケースに半永久的に保管し、数年後でも複数回再イメージングすることができます41。FE-SEMは、高解像度のイメージング、高倍率、汎用性を提供できるため、さまざまな研究分野でツールとして高く評価されています42。オルガネラの微細構造を3Dで表示するために、FE-SEM 43,44によって生成された後方散乱電子を使用して有用な解像度でシリアル2D画像スタックを生成する技術を使用して、特別な装置なしでターゲット領域またはそれらに関連する構造のハイスループットおよびマルチスケールイメージングを達成することもできます45.電荷アーチファクトの生成は、取得した画像の品質に直接影響するため、滞留時間を短くすることが特に重要です。
したがって、本研究では、ミトコンドリアクリステ46の3D構造を再構築するためにこのSEM技術で採用された実験手順について詳しく説明します。具体的には、従来のOTO標本作製法44,47を用いたスライスサンプルの作成、ウルトラミクロトームスライスによる切片採取の完了、FE-SEMによる逐次2Dデータの取得など、Amiraソフトウェアを用いたミトコンドリア領域の半自動セグメンテーションと3D再構成のデジタル化を実現するためのプロセスを紹介します。
