Hier beschrijven we een protocol voor de toepassing van high-throughput RNAi screening te ontdekken van de doelen van de gastheer die kunnen worden gemanipuleerd om het verbeteren van oncolytische virus therapie, specifiek rhabodvirus en vaccinia virus therapie, maar het kan gemakkelijk aangepast worden aan andere oncolytische virus platformen of voor het ontdekken van host-genen die virus replicatie in het algemeen moduleren.
