Het protocol stelt de Escherichia coli-selectieve druk opneming van niet-canonieke aminozuren (ncAAs) in het lactococcal antimicrobiële peptide nisine gebaseerd. De eigenschappen kunnen worden gewijzigd tijdens de recombinante expressie via substitutie met de gewenste ncAAs in de gedefinieerde groei media. Resulterende mutaties in de topicale worden toegewezen door groei remming testen en fluorescentie microscopie.
