Hier presenteren we een protocol voor het ontwikkelen van genetisch gemodificeerde muismodellen met behulp van embryonale stamcellen, vooral voor grote DNA knock-in (KI). Dit protocol is afgestemd met behulp van CRISPR / Cas9-genoombewerking, wat resulteert in een aanzienlijk verbeterde KI-efficiëntie in vergelijking met de conventionele homologe recombinatie-gemedieerde lineaire DNA-targetingmethode.
