Abstract

Introduction

Blandt alle stamceller med forskellige replikative kræfter, har dental stamceller som de postnatale stamceller henledt opmærksomheden i de senere år på grund af deres tilgængelighed, plasticitet og høj proliferativ evne i sammenligning med andre voksne stamceller 16. Karakteristisk lighed med de mesenkymale stamceller, dental pulp stamceller er adhærente klonogene celler, der har multiple differentiering kapacitet til mesenkym-og / eller ikke-mesenchym linier, både in vitro og in vivo. 1-8,17,18 DPSCs identificeres ved deres negative udtryk for hæmatopoietiske antigener (f.eks CD45, CD34, CD14), og positivt udtryk for CD90, CD29, CD73, CD105, CD44 og STRO-1. 19,20

Protocol

1. Forbered enzymopløsningen og spredning Medium (PM)

  1. Gør Collagenase Type I Løsning: Afvej collagenase type I (12 mg / ml) og opløses i 1 ml PBS og filtreres under anvendelse af en 0,2 um sprøjtefilter. Derefter placere det 15 ml rør og holde det ved -20 ° C indtil brug.

2. Forbered Humant Dental pulpavæv for Dental Pulp Stem Cell Isolation

  1. Normale humane tredje molarer blev indsamlet fra voksne (21-29 år) på Dental Clinic af Imam Ali under godkendte Institutional Review Board (IRB).
  2. Tænderne blev anbragt i det basale medium (α-MEM suppleret med 10% FBS) og blev overført til laboratoriet ved 4 ° C.
  3. Under sterile betingelser, i en biologisk laminar strømningshætte arbejder, set-up blev udført en 100 mm petriskåle for hver tand, der skal behandles. Tandoverflader blev renset med 70% ethanol.
  4. Mens du arbejder i en af ​​de petriskåle blev tand holdt ved dental pincet og en skalpel blev brugt til at rense ud snavs og parodontal ligament på rødderne.
  5. Cemento-emalje krydset blev langsomt skåret ved hjælp af steriliseret dental disk for at afsløre pulpakammeret. Det bør tages i betragtning, at opskæringen skal udføres langsomt for at reducere overophedning af den dentale væv.
  6. Pulpavæv blev forsigtigt adskilt fra kronen og roden. Pulpavæv blev skåret i små stykker ved hjælp af et skalpelblad.

3. Humant Dental Pulp Isolation

  1. Pulp væv undergik enten enzymatisk dissociation af papirmasse væv eller direkte celleudgroning fra pulpavæv eksplantater til isolering af hDPSCs.
  2. Enzymatisk dissociation af pulpavæv:
    1. Små stykker af papirmasse væv blev overført til enzymopløsning i 1 time ved 37 ° C. Vortex hver 30 min for at hjælpe med opbrydning væv.
    2. Derefter blev store celleaggregater fjernet, og enkeltcelle-suspensioner blev fremstillet ved at lede celler gennem en 70-pM cell strainer.
    3. Enkelt-cellesuspensioner blev centrifugeret ved 1200 rpm i 5 min ved stuetemperatur.

  3. Direkte celleudgroning fra pulpavæv eksplantater:
    1. Pulpavæv blev hakket i 1-2 mm stykker, og hvert stykke blev anbragt i 25-cm2 dyrkningskolber med PM og derefter inkuberet ved 37 ° C i 5% CO2.     3. De vokset celler ved konfluens blev sub-dyrket ved forhold på 1:4 (passage 1).

4. Immunophenotyping

  1. 250.000 celler /rør af begge typer celler i passage 3 blev inkuberet med PE eller FITC-konjugerede anti-humane antistoffer med deres isotyper i 30 minutter ved 4 ° C i mørke. (Bemærk:. Koncentrationen af antistoffer blev anvendt i overensstemmelse med producentens protokol)
  2. Efter inkubationstiden blev 100 pi PBS eller FACS fortynding opløsning sættes til hvert rør og centrifugeret ved 1200 rpm i 5 minutter i mørke.
  3. Supernatanterne blev fjernet og pellets resuspenderet i 100 pi PBS eller FACS fortynding i mørke.
  4. Målrettede overflademarkører (CD markører) blev evalueret ved BD FACSCalibur.

5. Induktion af differentiering af DPSCs i mineraliseret Cells & Kvantificeret Alizarin red S Assay

  1. DPSCs, som blev opnået ved enten enzymatisk dissociation af pulpavæv (kendt som dpsc-ED) eller direkte celleudgroning fra pulpavæv eksplantater (dpsc-OG) var sub-dyrket i 3 passager.
  2. At passage 3, blev celler frø i en 6 brønds plade ved 5.000 celler / brønd.
  3. Ved konfluens på 60%, blev odontogen medium erstattet med konventionelle PM. Tre brønde blev holdt som negativ kontrol ved tilsætning af vækstmedium.
  4. Medium ændre hver 3 dage, indtil dag 21.
  5. På dag 21, blev cellerne vasket med PBS og blev fikseret med 1 ml / brønd 10% paraformaldehyd i 15 minutter ved stuetemperatur.
  6. Derefter paraformaldehyd blev pipetteret fra, omhyggeligt og cellerne blev vasket 3 gange (5-10 min hver gang) med destilleret H 2 O. (Bemærk: Undgå at forstyrre monolag.)
  7. Efter vask blev 1 ml / brønd Alizarin red S tilsat i 20 minutter ved stuetemperatur.
  8. Celler blev vasket med deioniseret H 2 O fire gange (5 minutter hver gang med forsigtig vipning) for at fjerne eventuelle yderligere Alizarin red.
  9. 1 ml / brønd H2O blev tilsat for at forhindre cellerne i at tørre.
  10. Pladerne blev antaget til visuelle inspection og / eller billede erhvervelse.
  11. For Alizarin Red S kvantificering assay blev H2O erstattet med 1 ml 10% eddikesyre til hver brønd på 6-brønds plade og inkuberes i 30 min ved stuetemperatur med omrystning.
  12. Derefter cellemonolaget / brønd blev skrabe & både eddikesyre og celler blev overført til separate 1,5 ml mikro-centrifugerør. (Bemærk:. Hver brønd skal være overførsel til et separat rør hvilket er tre testbrønde & tre kontrolgrupper brønde for hver ED & OG grupper)
  13. Rørene blev hvirvelbehandlet kraftigt i 30 sek.
  14. Der opvarmes til 85 ° C i 10 minutter. (Bemærk: For at undgå fordampning blev rørene forseglet med parafilm.)
  15. Efter opvarmning blev rørene overført til is i 5 minutter. (Bemærk: Rør ikke bør åbnes før blive afkølet.)
  16. Opslæmningerne blev centrifugeret ved 20.000 x g i 15 minutter. Mens centrifugering blev Alizarin red standarder træffesop.

Alizarin red S standard blev fremstillet ved først at fortynde fortyndingspufferen og anvendes til fremstilling af to-fold seriefortyndinger baseret på bordet.

  1. Efter centrifugering blev 1 ml supernatant / rør overført til nye separate 1,5 ml mikro-centrifugerør.
  2. PH-værdien blev neutraliseret med ~ 375 | jl 10% Ammoniumhydroxid per rør. (Bemærk: pH rage bør være 4,1-4,5.)

Representative Results

  1. DPSCs, der blev opnået ved enzymatisk dissociation (dpsc-ED) er vist her på dag 10, 15,18 (fig. 1). Kolonierne indledt til dannelse på næsten 3-5 dage efter isolering.
  2. Vokset DPSCs (dpsc-OG) er vist i figur 2 på dag 5, 10, 13 & 18. Fibroblastlignende celler begyndte at migrere fra pulpavæv til kolben ved parallelle bestilling med næsten 5 dage efter podning.
  3. DPSCs ved passage 3 ved hjælp af begge fremgangsmåder er vist i figur 3.. Begge typer DPSCs var næsten i samme størrelse og morfologi.
  4. Immunophenotyping resulterer i dpsc-ED og dpsc-OG (fig. 4). Disse resultater indikerede tilstedeværelsen af mesenchymale stamceller er markører, såsom CD44, CD73, CD105 og CD90 og fraværet af hæmatopoietiske & endothelial markører, såsom CD34, CD45 og CD11b . De udtryk for CD105 og CD146 (perivaskulær markør) var mere i dpsc-OG sammenlignet med dpsc-ED.STRO-1 var negativ i begge grupper.   5. Alizarin red S morfologi og kvantificering resultater for evaluering af mineraliseret væv dannelse i dpsc-ED og dpsc-OG på dag 21 er vist i fig. 5. Discussion

Denne protokol beskriver processen med isolering og karakterisering af hDPSCs fra dental pulp ved hjælp af to metoder, enzymatisk dissociation og direkte udvækst af stamceller fra pulpavæv eksplantater. Desuden, in vitro differentiering af disse celler i odontoblasts blev vurderet ved kvantitativ Alizarin Red S assay og qPCR.

Eksisterende protokoller til isolering pulpavæv fra human tand var blevet brugt forskellige instrumenter, såsom tang (knogle tang) 9, udryddelse nål 29, Gracey curette 30, dental revne bor 4, osv. Disse metoder er tidskrævende, samtidig overveje lavt niveau overophedning af papirmasse ved tilsætning af vand kontinuerligt. I den nuværende teknik, er dental diamantskive anvendes til isolering af papirmasse væv, hvilket er en hurtig måde at skære tænder med minimal kontaminering i ettrinsproces, og minimerer overophedning af pulp under en skæreproces. Desuden hDPSCs enre endeligt differentierede til odontoblasts som er aldrig sammenlignet i forbindelse med humane blivende tænder ved diskuteret formål. Sammenlignende vurdering af differentierede hDPSCs (ED & OG grupper) ved kvantitativ Alizarin red S assay indikerede højere mineralisering potentiale hDPSC-ED i sammenligning med OL gruppe. Disse resultater bekræftes også af qPCR. DSPP, ALP og MEPE som mineralisering markører opreguleret efter odontoblast differentiering i begge grupper. På den anden side var udtryk for MEPE & ALP højere i opdelte hDPSC-ED sammenlignet med kontrollen. Efter aftale med tidligere undersøgelse om løvfældende tænder 27, også stamceller fra blivende tænder ved hjælp af ED eller OL isoleringsmetoder udviser lignende differentiering adfærd under de samme betingelser. Desuden, for bedre sammenlignende evaluering, i denne undersøgelse, var begge hDPSC-ED & hDPSC-OG isoleret fra de samme individer.

Udtryk for MSC markører end fravær af heamatopietic / endothelial markører identificerer begge typer hDPSCs som mesenkymale stamceller. På den anden side var CD 105 / CD 146 højere i dpsc-OG sammenlignet med ED grupper. Trods de nuværende bevis efter tilstedeværelsen af STRO-1 i DPSCs 31, viste vores resultater ingen ekspression af STRO-1 i begge grupper. Kan imidlertid betydelige forskelle i overflademarkør ekspression i forskellige undersøgelser skyldes de enten dyrkningsbetingelserne såsom passage nummer, sammensætning af medier osv. eller inter-individuelle variationer mellem forskellige donorer. Fraværet af STRO-1 i begge grupper kan indikere, at ekspression af denne markør måske ikke direkte påvirke odontoblast differentieringspotentiale (ikke vist i video).

Forskellige hDPSCs isolation tilstand kan bringe anderledes differentiering kapacitet. Disse forskelle kan skyldes, at eksistensen af ​​forskellige befolkningsgrupper i Pulpavævet. Således vælger isolation metoder MIGht være nyttige til målrettet-vævsspecifikke reparation & regeneration for fremtidige kliniske tilgange. Vores resultater indikerede højere mineralisering kapacitet enzymatisk fordøjet dpsc forhold til outgrown dem. Der er dog yderligere undersøgelser, såsom vurderingen af dannelsen af rørformede struktur in vivo og in vitro, der kræves for at afgøre, hvilken af disse to procedurer er gældende for funktionel dentin / papirmasse vævsregeneration behandlinger.
