Dit protocol beschrijft de afleiding van transplanteerbare hematopoietische stamcellen uit muis embryonale stamcellen (ESC) en de daaropvolgende injectie in dodelijk bestraalde ontvangende muizen. In het kort, worden ESC gedifferentieerd embryoid lichamen, die vervolgens besmet zijn met retrovirale HoxB4 en co-gekweekt met OP9 stromale cellen en hematopoietische cytokines.
