Vi beskriver en liten molekyl-baserat protokoll för differentiering av mus-embryonala stamceller till oligodendrocyte föregångare celler (OPC). Detta protokoll genererar Olig2 + NG2 + OPCs med hög effektivitet med 30 dagar av differentiering. Vi beskriver också en metod att generera "tillsatta" OPCs som kan avfyra aktionspotentialer.
