Memory retrieval initiates two consolidation processes: consolidation of an extinction memory and reconsolidation of the acquisition memory. The strength of the consolidation processes depends on both the strength of the acquisition memory and the strength of retrieval trials and is correlated with its sensitivity to inhibition. We demonstrate that in the honeybee (Apis mellifera), memory retrieval of a consolidated appetitive olfactory memory leads to both consolidation processes, depending on the number of retrieval trials. Spontaneous recovery from extinction is induced by many (five), but not by few (one and two), retrieval trials. Spontaneous recovery is blocked by emetine, an inhibitor of protein synthesis. We conclude that reconsolidation of the acquisition memory underlies spontaneous recovery.

R&#xE9;cup&#xE9;ration spontan&#xE9;e est bloqu&#xE9;e par l&#x26;#39;&#xE9;m&#xE9;tine, un inhibiteur de la synth&#xE8;se des prot&#xE9;ines. Memoria recupero avvia due processi di consolidamento: consolidamento di un&#39;estinzione memoria e organico della memoria di acquisizione. La forza dei processi di consolidamento dipende la forza della memoria di acquisizione sia la forza delle prove di recupero ed &egrave; correlata con la sua sensibilit&agrave; all&#39;inibizione. Dimostriamo che nell&#39;APE (Apis mellifera), recupero della memoria di una memoria olfattiva appetitiva consolidato conduce a entrambi i processi di consolidamento, a seconda del numero delle prove di recupero. Retrieving a consolidated memory--by exposing an animal to the learned stimulus but not to the associated reinforcement--leads to two opposing processes: one that weakens the old memory as a result of extinction learning, and another that strengthens the old, already-consolidated memory as a result of some less well-understood form of learning. This latter process of memory strengthening is often referred to as "reconsolidation", since protein synthesis can inhibit this form of memory formation. Although the behavioral phenomena of the two antagonizing forms of learning are well documented, the mechanisms behind the corresponding processes of memory formation are still quite controversial. Referring to results of extinction/reconsolidation experiments in honeybees, we argue that two opposing learning processes--with their respective consolidation phases and memories--are initiated by retrieval trials: extinction learning and reminder learning, the latter leading to the phenomenon of spontaneous recovery from extinction, a process that can be blocked with protein synthesis inhibition.

Abrufen eines konsolidierten Speichers--durch Verf&uuml;gbarmachen eines Tieres auf den gelernten Reiz, aber nicht auf die damit verbundenen Verst&auml;rkung--f&uuml;hrt zu zwei gegens&auml;tzlichen Prozesse: einen mit den alten Speicher durch Aussterben lernen schw&auml;cht, und einen, der den alten, bereits konsolidierten Speicher aufgrund einiger weniger verst&auml;ndliche Form des Lernens st&auml;rkt. Dieser letzteren Proze&szlig; Speicher zu st&auml;rken wird oft als &quot;Reconsolidation&quot;, bezeichnet, da diese Form der Ged&auml;chtnisbildung zu Proteinsynthese hemmen kann. Obwohl die verhaltensbasierte Ph&auml;nomene der zwei Nguyens Formen des Lernens gut dokumentiert sind, sind die Mechanismen hinter den entsprechenden Prozessen der Ged&auml;chtnisbildung noch sehr umstritten. Unter Bezugnahme auf Ergebnisse der Honigbienen Aussterben/Reconsolidation Experimente, wir argumentieren, dass zwei gegnerische Lernprozesse--mit ihren jeweiligen Konsolidierung Phasen und Erinnerungen--durch Abruf Studien initiiert werden: Aussterben lernen und Erinnerung lernen, das letztere f&uuml;hrende auf das Ph&auml;nomen der spontanen Erholung vor dem Aussterben, ein Prozess, der mit Protein-Synthese-Hemmung blockiert werden kann.

Combining memory retrieval with the application of a protein synthesis-inhibitor leads to an amnestic effect that is referred to as the reconsolidation phenomenon. Several behavioural studies demonstrate that only a few or weak retrieval trials (that do not result in significant extinction) lead to this phenomenon. In contrast, many trials (that result in significant extinction) combined with a protein synthesis inhibitor result in an inhibition of the extinction memory. Based on these findings it was suggested that extinction is the boundary condition for reconsolidation: when extinction is induced the consolidation of the extinction memory is the dominant process. Recently we were not able to confirm this hypothesis in the honeybee (Apis mellifera): we did not find the reconsolidation phenomenon after one retrieval trial, but demonstrated reconsolidation after five retrieval trials that led to extinction. To exclude that this observation resembles a special case in insects we here wanted to know if one retrieval trial induces reconsolidation as it has been demonstrated before in many other species. To do so we used experimental parameters that had been used before to demonstrate consolidation in the honeybee with the exception that this time the protein synthesis-inhibitor was applied 1 h after one memory retrieval instead after acquisition. We thereby demonstrate the reconsolidation phenomenon after one retrieval trial but only when using the doubled dose of protein synthesis-inhibitor that has been used to inhibit consolidation.

Pour exclure que cette observation ressemble &#xE0; un cas particulier chez les insectes que nous ici a voulu savoir si un essai de r&#xE9;cup&#xE9;ration induit reconsolidation comme il a &#xE9;t&#xE9; d&#xE9;montr&#xE9; auparavant dans de nombreuses autres esp&#xE8;ces. Pour ce faire, nous avons utilis&#xE9; les param&#xE8;tres exp&#xE9;rimentaux qui avaient &#xE9;t&#xE9; utilis&#xE9;s avant pour d&#xE9;montrer la consolidation de l&#x26;#39;abeille &#xE0; l&#x26;#39;exception que cette fois la synth&#xE8;se des prot&#xE9;ines-inhibiteur a &#xE9;t&#xE9; appliqu&#xE9;e 1 h apr&#xE8;s une r&#xE9;cup&#xE9;ration de la m&#xE9;moire au lieu apr&#xE8;s l&#x26;#39;acquisition. La combinaci&#xF3;n de recuperaci&#xF3;n de la memoria con la aplicaci&#xF3;n de una s&#xED;ntesis de prote&#xED;nas-inhibidor conduce a un efecto amn&#xE9;sico que se conoce como el fen&#xF3;meno de reconsolidaci&#xF3;n. Varios estudios sobre el comportamiento demuestran que s&#xF3;lo unos pocos o los ensayos de recuperaci&#xF3;n d&#xE9;bil (que no dan lugar a la extinci&#xF3;n significativo) dar lugar a este fen&#xF3;meno. En contraste, muchos ensayos (que resultan en la extinci&#xF3;n significativo) combinado con un inhibidor de la s&#xED;ntesis de prote&#xED;nas resultado en una inhibici&#xF3;n de la memoria extinci&#xF3;n. Bas&#xE1;ndose en estos hallazgos se sugiri&#xF3; que la extinci&#xF3;n es la condici&#xF3;n l&#xED;mite para la reconsolidaci&#xF3;n: cuando la extinci&#xF3;n se induce la consolidaci&#xF3;n de la memoria de la extinci&#xF3;n es el proceso dominante. Recientemente no hemos podido confirmar esta hip&#xF3;tesis en la abeja mel&#xED;fera (Apis mellifera): no encontramos el fen&#xF3;meno de reconsolidaci&#xF3;n despu&#xE9;s de un juicio de recuperaci&#xF3;n, pero demostr&#xF3; la reconsolidaci&#xF3;n despu&#xE9;s de cinco ensayos de recuperaci&#xF3;n que llevaron a la extinci&#xF3;n. Para excluir que esta observaci&#xF3;n se asemeja a un caso especial en los insectos que aqu&#xED; quer&#xED;a saber si un ensayo de recuperaci&#xF3;n induce la reconsolidaci&#xF3;n como se ha demostrado antes en muchas otras especies. Para ello se utilizaron los par&#xE1;metros experimentales que se hab&#xED;an utilizado antes para demostrar la consolidaci&#xF3;n de la abeja con la excepci&#xF3;n de que esta vez el inhibidor de la s&#xED;ntesis de prote&#xED;nas-se aplic&#xF3; 1 h despu&#xE9;s de una recuperaci&#xF3;n de la memoria en lugar de despu&#xE9;s de la adquisici&#xF3;n. Nosotros as&#xED; demostrar el fen&#xF3;meno de reconsolidaci&#xF3;n despu&#xE9;s de ensayo de recuperaci&#xF3;n de un pero s&#xF3;lo cuando se usa la dosis duplicado de la s&#xED;ntesis de prote&#xED;nas-inhibidor que se ha utilizado para inhibir la consolidaci&#xF3;n.

Un ensayo de recuperaci&#xF3;n induce reconsolidaci&#xF3;n en un paradigma de aprendizaje apetitivo en las abejas (Apis mellifera)

Here, we examine the role of the magnitude of the unconditioned stimulus (US) during classical conditioning in consolidation processes after memory retrieval. We varied the US durations during training and we test the impact of these variations on consolidation after memory retrieval with one or two conditioned stimulus-only trials. We found that the consolidation of an extinction memory depends on US duration during training and ruled out the possibility that this effect is attributable to differences in satiation after conditioning. We conclude that consolidation of an extinction memory is triggered only when the duration of the US reaches a critical threshold. Memory formation is a continuous process composed of multiple phases that can develop independently from each other. These phases depend on signaling pathways initiated after the activation of receptors in different brain regions. The NMDA receptor acts as a sensor of coincident activity between neural inputs, and, as such, its activation during learning is thought to be crucial for various forms of memory. In this study, we inhibited the expression of the NR1 subunit of the NMDA receptor in the honeybee brain using RNA interference. We show that the disruption of the subunit expression in the mushroom body region of the honeybee brain during and shortly after appetitive learning selectively impaired memory. Although the formation of mid-term memory and early long-term memory was impaired, late long-term memory was left intact. This indicates that late long-term memory formation differs in its dependence on NMDA receptor activity from earlier memory phases.

In dieser Studie hemmte wir Ausdruck der NR1-Untereinheit des NMDA-Rezeptors im Gehirn Honigbiene mithilfe der RNA-Interferenz. Wir zeigen, dass die St&ouml;rung des Ausdrucks Untereinheit in der Pilz-K&ouml;rper-Region des Gehirns Honigbiene w&auml;hrend und kurz nach Vorabend lernen gezielt Ged&auml;chtnisst&ouml;rungen. Obwohl die Bildung von Mid-Term Memory und fr&uuml;hen Langzeitged&auml;chtnis beeintr&auml;chtigt wurde, war sp&auml;t Langzeitged&auml;chtnis intakt. Dies bedeutet, dass die sp&auml;ten langfristige Ged&auml;chtnisbildung in seine Abh&auml;ngigkeit von NMDA-Rezeptor-Aktivit&auml;t von fr&uuml;her Speicher Phasen unterscheidet.

Conditioned behavior as observed during classical conditioning in a group of identically treated animals provides insights into the physiological process of learning and memory formation. However, several studies in vertebrates found a remarkable difference between the group-average behavioral performance and the behavioral characteristics of individual animals. Here, we analyzed a large number of data (1640 animals) on olfactory conditioning in the honeybee (Apis mellifera). The data acquired during absolute and differential classical conditioning differed with respect to the number of conditioning trials, the conditioned odors, the intertrial intervals, and the time of retention tests. We further investigated data in which animals were tested for spontaneous recovery from extinction. In all data sets we found that the gradually increasing group-average learning curve did not adequately represent the behavior of individual animals. Individual behavior was characterized by a rapid and stable acquisition of the conditioned response (CR), as well as by a rapid and stable cessation of the CR following unrewarded stimuli. In addition, we present and evaluate different model hypotheses on how honeybees form associations during classical conditioning by implementing a gradual learning process on the one hand and an all-or-none learning process on the other hand. In summary, our findings advise that individual behavior should be recognized as a meaningful predictor for the internal state of a honeybee--irrespective of the group-average behavioral performance.

Le comportement individuel a &#xE9;t&#xE9; caract&#xE9;ris&#xE9; par une acquisition rapide et stable de la r&#xE9;ponse conditionn&#xE9;e (RC), ainsi que par un arr&#xEA;t rapide et stable des stimuli suivants CR r&#xE9;compens&#xE9;es. Konditionierte Verhalten als beobachteten w&auml;hrend Klassische Konditionierung in einer Gruppe identisch behandelter Tiere gibt Einblicke in den physiologischen Prozess lernen und Ged&auml;chtnis-Formation. Mehrere Studien bei Wirbeltieren fand jedoch einen bemerkenswerten Unterschied zwischen der Konzerndurchschnitt Verhaltens-Leistung und die Verhaltensmerkmale einzelner Tiere. Hier analysieren wir eine gro&szlig;e Anzahl von Daten (1640 Tiere) auf olfaktorische Konditionierung in die Honigbiene (Apis Mellifera). Die Daten, die w&auml;hrend der absoluten und differenzielle Klassische Konditionierung erworben unterschieden sich in Bezug auf die Anzahl der Konditionierung Studien, die bedingte Ger&uuml;che, die intertrial Intervalle und die Zeit der Aufbewahrung Pr&uuml;fungen. Wir untersuchten weitere Daten, die bei denen Tiere f&uuml;r spontane Erholung vor dem Aussterben getestet wurden. In allen Datens&auml;tzen fanden wir, dass die allm&auml;hlich zunehmende Konzerndurchschnitt Lernkurve nicht angemessen das Verhalten einzelner Tiere dar. Individuelles Verhalten zeichnete sich durch eine schnelle und stabile Akquisition der konditionierten Reaktion (CR) sowie durch eine schnelle und stabile Einstellung unbelohnt Reize nach CR. Dar&uuml;ber hinaus haben wir pr&auml;sentieren und anderes Modell Hypothesen &uuml;ber wie Honigbienen Verb&auml;nde w&auml;hrend Klassische Konditionierung bilden, durch die Implementierung einer schrittweisen Lernprozess einerseits und ein termingebundener Lernprozess auf der anderen Seite zu bewerten. Zusammenfassend l&auml;sst sich sagen beraten unsere Ergebnisse, dass individuelles Verhalten als eine sinnvolle Pr&auml;diktor f&uuml;r den internen Zustand des einer Honigbiene--unabh&auml;ngig von der Gruppe &uuml;berdurchschnittlich verhaltensbasierte Leistung erkannt werden sollen.

Comportamento condizionato come osservato durante il condizionamento classico in un gruppo di animali trattati in modo identico fornisce intuizioni il processo fisiologico di formazione apprendimento e memoria. Tuttavia, diversi studi nei vertebrati trovano una notevole differenza tra le prestazioni comportamentali di gruppo-media e le caratteristiche del comportamento dei singoli animali. Sin embargo, varios estudios en los vertebrados se encontr&#xF3; una diferencia notable entre el rendimiento del grupo de la media del comportamiento y las caracter&#xED;sticas de comportamiento de los animales individuales. En este caso, hemos analizado una gran cantidad de datos (1640 animales) en el condicionamiento olfativo en la abeja mel&#xED;fera (Apis mellifera). Los datos adquiridos durante el condicionamiento cl&#xE1;sico, absoluta y diferencial difieren con respecto al n&#xFA;mero de ensayos de condicionamiento, los olores acondicionado, los intervalos entre ensayos, y el tiempo de las pruebas de retenci&#xF3;n. Adem&#xE1;s, investig&#xF3; los datos en la que los animales se pusieron a prueba para la recuperaci&#xF3;n espont&#xE1;nea de la extinci&#xF3;n. En todos los conjuntos de datos se encontr&#xF3; que el aumento gradual del grupo a la media curva de aprendizaje no representan adecuadamente el comportamiento de los animales individuales. El comportamiento individual se caracteriza por una r&#xE1;pida adquisici&#xF3;n y estable de la respuesta condicionada (RC), as&#xED; como por un cese r&#xE1;pido y estable de los est&#xED;mulos siguientes CR recompensa. Adem&#xE1;s, presentar y evaluar diferentes hip&#xF3;tesis de modelo sobre c&#xF3;mo las abejas formar asociaciones en el condicionamiento cl&#xE1;sico mediante la implementaci&#xF3;n de un proceso gradual de aprendizaje, por un lado y un proceso de aprendizaje de todo o ninguno en la otra mano. En resumen, nuestros resultados recomendamos que el comportamiento individual debe ser reconocido como un predictor significativo para el estado interno de una abeja - independientemente de los resultados del comportamiento del grupo de la media.
