We previously reported that the Rho-Rho kinase pathway controls cyclin D1 expression by preventing its early G1 phase induction in response to Rac and/or Cdc42, thus increasing its dependence on ERK signaling and actin stress fiber formation. We now show that the Rho kinase effector LIM kinase is responsible for this effect. Surprisingly, inhibition of Rac-dependent cyclin D1 expression by LIM kinase is independent of both cofilin phosphorylation and actin polymerization. Instead, specific mutation of its nuclear localization and export sequences showed that LIM kinase acts in the nucleus to suppress Rac/Cdc42-dependent cyclin D1 expression. Our results therefore describe an unexpected role for LIM kinase that requires nuclear translocation. The effect of nuclear LIM kinase on cyclin D1 expression ultimately regulates the duration of G1 phase and the degree to which G1 phase progression depends on actin stress fiber formation and imposition of cellular tension.

Abbiamo segnalato in precedenza che i Rho-Rho kinase pathway controlli ciclina D1 espressione prevenendo la sua induzione di fase G1 precoce in risposta a Rac e/o Cdc42, aumentando cos&igrave; la sua dipendenza da ERK segnalazione e actina stress formazione di fibra. In yeast, the transcriptional adaptor yeast Ada2 (yAda2) is a part of the multicomponent SAGA complex, which possesses histone acetyltransferase activity through action of the yGcn5 catalytic enzyme. yAda2, among several SAGA proteins, serves to recruit SAGA to genes via interactions with promoter-bound transcription factors. Here we report identification of a new human Ada2 homologue, hAda2beta. Ada2beta differs both biochemically and functionally from the previously characterized hAda2alpha, which is a stable component of the human PCAF (human Gcn5 homologue) acetylase complex. Ada2beta, relative to Ada2alpha, interacted selectively, although not stably, with the Gcn5-containing histone acetylation complex TFTC/STAGA. In addition, Ada2beta interacted with Baf57 (a component of the human Swi/Snf complex) in a yeast two-hybrid screen and associated with human Swi/Snf in vitro. In functional assays, hAda2beta (but not Ada2alpha), working in concert with Gcn5 (but not PCAF) or Brg1 (the catalytic component of hSwi/Snf complex), increased transcription via the B-cell-specific transcription factor Pax5/BSAP. These findings support the view that Gcn5 and PCAF have distinct roles in vivo and suggest a new mechanism of coactivator function, in which a single adaptor protein (Ada2beta) can coordinate targeting of both histone acetylation and chromatin remodeling activities.

Aqu&iacute; divulgamos la identificaci&oacute;n de un nuevo hom&oacute;logo humano de Ada2, hAda2beta. Ada2beta difiere tanto bioqu&iacute;mico y funcionalmente la hAda2alpha previamente caracterizados, que es un componente estable de la humana PCAF (hom&oacute;logo humano Gcn5) acetilasa complejo. Ada2beta, en relaci&oacute;n con Ada2alpha, interactuaron selectivamente, aunque no estable, con la acetilaci&oacute;n de histonas que contienen Gcn5 complejo TFTC/STAGA. Adem&aacute;s, Ada2beta interactu&oacute; con Baf57 (un componente de la humana Swi/Snf complejo) en una pantalla de dos h&iacute;bridos de levadura y asociado humano Swi/Snf in vitro. p21(cip1) inhibits S phase entry by binding to cyclin-cdk2 (cyclin-dependent kinase-2) complexes. The levels of p21(cip1) are rapidly induced after mitogenic stimulation of quiescent fibroblasts and then down-regulate as the cells reach late G(1) phase and activate cyclin E-cdk2. In this study, we have shown that pharmacological inhibition of protein kinase C (PKC), expression of dominant negative PKCdelta, or knockdown of PKCdelta with small interfering RNA elevates p21(cip1) protein levels in mouse embryo fibroblasts. This effect is selective, post-transcriptional, and proteasome-dependent but distinct from previously identified post-transcriptional control mechanisms involving cyclin D1 and Skp2. PKCdelta inhibition results in a reduced entry into S phase, and this effect is not detected in p21(cip1)-null cells. Thus, post-transcriptional destabilization of p21(cip1) appears to be a major mitogenic effect of PKCdelta in fibroblasts.

Cooperative signaling between growth factor receptor tyrosine kinases, integrins, and the actin cytoskeleton is required for activation of the G1-phase cyclin-dependent kinases and progression through G1-phase. Increasing evidence suggests that there is cell type specificity in these cooperative interactions and that the compliance of the underlying substratum can strongly affect adhesion-dependent signaling to the cell cycle. This chapter reviews our current methods for studying how cell type specificity and changes in substratum compliance can contribute to G1-phase cell cycle control. We also describe several of our current analytical procedures.

Integrin-mediated adhesion to substratum is required for cyclin D1 induction in mesenchymal cells, but we show here that the induction of cyclin D1 persists despite blockade of ECM-integrin signaling in MCF10A mammary epithelial cells. E-cadherin-mediated cell-cell adhesion also supports cyclin D1 induction in these cells, and the combined inhibition of both E-cadherin and integrin adhesion is required to prevent the expression of cyclin D1 mRNA and protein. Our previous studies described a pro-proliferative effect of E-cadherin in MCF10A cells, mediated by Rac, and we now show that Rac is required for cyclin D1 mRNA induction by both E-cadherin and integrin engagement. The levels of p21Cip1 and p27Kip1, Cdk inhibitors that are also targets of integrin signaling, are not affected by E-cadherin-mediated cell-cell adhesion. Finally, we show that the increased expression of cyclin D1 mRNA associated with E-cadherin-dependent cell-cell adhesion is causally linked to an increased entry into S phase. Our results identify Rac signaling to cyclin D1 as a crucial pro-proliferative effect of E-cadherin-mediated cell-cell adhesion.

We describe here the regulation of ABCG2 expression and side population (SP) abundance in MCF7 human breast cancer cells. The level of ABCG2 mRNA and protein were increased in purified MCF7 SP relative to non-SP cells, and incubation with an ABCG2-specific inhibitor or ABCG2 short interfering RNA eliminated the MCF7 SP. The purified MCF7 SP could generate a heterogeneous population containing both SP and non-SP cells in culture. In vivo tumorigenicity experiments showed that the purified MCF7 SP has an increased ability to colonize the mouse mammary gland. Importantly, the MCF7 SP was depleted by a transforming growth factor-beta (TGFbeta)-directed epithelial-mesenchymal transition (EMT), and this effect was associated with a strong down-regulation of ABCG2 gene expression, and an increased sensitivity to mitoxantrone. ABCG2 expression and SP abundance were restored upon the removal of transforming growth factor-beta and reversion of the cells to an epithelial phenotype. Knock-down of E-cadherin also reduced SP abundance, but this effect was not accompanied by the loss of ABCG2 mRNA or protein. We conclude that ABCG2 expression in MCF7 cells is regulated during an EMT, and that the EMT effect reflects posttranslational regulation of ABCG2 function by E-cadherin as well as transcriptional repression of the ABCG2 gene.

Le niveau de ABCG2 ARNm et la prot&#xE9;ine purifi&#xE9;e a &#xE9;t&#xE9; augment&#xE9; dans MCF7 SP par rapport aux non-SP cellules, et l&#x26;#39;incubation avec un inhibiteur de ABCG2-sp&#xE9;cifique ou ABCG2 courts ARN interf&#xE9;rents &#xE9;limin&#xE9; le SP MCF7. Il livello della ABCG2 mRNA e proteina sono stati aumentati in SP MCF7 purificato rispetto le cellule non-SP e incubazione con un inibitore specifico ABCG2 o RNA interferente breve ABCG2 eliminato MCF7 SP. La SP MCF7 purificato potrebbe generare una popolazione eterogenea contenenti sia SP e SP non cellule in coltura. El nivel de ABCG2 ARNm y prote&#xED;na se increment&#xF3; en purific&#xF3; MCF7 SP relativa a la no-SP c&#xE9;lulas, y la incubaci&#xF3;n con un inhibidor espec&#xED;fico de ABCG2 o ABCG2 ARN corto de interferencia eliminado el SP MCF7. Stents eluting anti-proliferative drugs limit restenosis, but drugs commonly used to date are relatively non-specific cytostatic agents which inhibit proliferation of intimal endothelial cells as well as medial smooth muscle cells and may thereby contribute to the clinical complications associated with angioplasty. In an effort to identify a more specific anti-proliferative agent, we compared the effects of rapamycin to those of cicaprost, a mimetic of the naturally occurring anti-mitogen, PGI(2). Rapamycin and cicaprost were both strongly anti-mitogenic in vascular smooth muscle cells (VSMCs). But unlike rapamycin, cicaprost did not inhibit mitogenesis in aortic endothelial cells even when used at concentrations >10-fold higher than its ED(50) for VSMCs. Similarly, both rapamycin and cicaprost have been reported to regulate levels of the cdk inhibitor, p27(kip1). But rapamycin remained anti-mitogenic in p27(kip1)-null VSMCs whereas the anti-mitogenic effect of cicaprost was completely dependent on p27(kip1). We conclude that stable PGI(2) mimetics may be highly specific inhibitors of p27(kip1)-dependent VSMC proliferation after vascular injury.

Cell-type- and cell-cycle-specific anti-mitogenesis by cicaprost.

Les stents &#xE0; &#xE9;lution de m&#xE9;dicaments anti-prolif&#xE9;ratifs de limiter la rest&#xE9;nose, mais les m&#xE9;dicaments couramment utilis&#xE9;s &#xE0; ce jour sont relativement non sp&#xE9;cifiques des agents cytostatiques qui inhibent la prolif&#xE9;ration des cellules endoth&#xE9;liales intimales ainsi que m&#xE9;dianes des cellules musculaires lisses et peuvent ainsi contribuer &#xE0; des complications cliniques associ&#xE9;es &#xE0; une angioplastie. Dans un effort pour identifier un plus sp&#xE9;cifique anti-prolif&#xE9;rative agent, nous avons compar&#xE9; les effets de la rapamycine &#xE0; ceux de cicaprost, un mim&#xE9;tique de l&#x26;#39;origine naturelle anti-mitog&#xE8;ne, IGP (2). La rapamycine et cicaprost &#xE9;taient tous les deux fortement anti-mitog&#xE9;nique dans les cellules musculaires lisses vasculaires (CMLV). Mais contrairement &#xE0; la rapamycine, cicaprost n&#x26;#39;a pas inhib&#xE9; la mitogen&#xE8;se dans les cellules endoth&#xE9;liales aortiques m&#xEA;me lorsqu&#x26;#39;ils sont utilis&#xE9;s &#xE0; des concentrations&#x26;gt; 10 fois plus &#xE9;lev&#xE9;e que l&#x26;#39;ES (50) pour les CMLV. De m&#xEA;me, &#xE0; la fois de la rapamycine et cicaprost ont &#xE9;t&#xE9; signal&#xE9;s &#xE0; r&#xE9;guler les niveaux de l&#x26;#39;inhibiteur de cdk, p27 (Kip1). Mais la rapamycine rest&#xE9;s anti-mitog&#xE9;nique dans p27 (Kip1)-nulles CMLV alors que l&#x26;#39;effet anti-mitog&#xE8;ne de cicaprost &#xE9;tait compl&#xE8;tement d&#xE9;pendante de p27 (Kip1). Nous concluons que la stabilit&#xE9; des IGP (2) mim&#xE9;tiques peuvent &#xEA;tre inhibiteurs hautement sp&#xE9;cifiques de p27 (Kip1)-d&#xE9;pendante la prolif&#xE9;ration des CMLV apr&#xE8;s une l&#xE9;sion vasculaire.

In dem Bem&uuml;hen, einen spezifischeren Anti-proliferativen Agent identifizieren verglichen wir die Auswirkungen der angiomiolipomas des Cicaprost, einer mimetischen des nat&uuml;rlich vorkommenden Anti-mitogen, PGI(2). Angiomiolipomas und Cicaprost waren beide stark anti-mitogenic in vaskul&auml;ren glatten Muskelzellen (VSMCs). Aber im Gegensatz zu angiomiolipomas, Cicaprost Mitogenesis in Aorten Endothelzellen, selbst wenn in Konzentrationen verwendet nicht hemmen &gt; 10-fache h&ouml;her als seine ED(50) f&uuml;r VSMCs. Ebenso angiomiolipomas und Cicaprost wurden beschrieben, Ebenen der Cdk-Inhibitor zu regulieren p27(kip1). Aber angiomiolipomas blieb anti-mitogenic in p27 (kip1)-VSMCs null, w&auml;hrend die anti-mitogenic Wirkung der Cicaprost vollst&auml;ndig von der p27(kip1) abh&auml;ngig war. Wir schlie&szlig;en, dass stabile PGI(2) Mimetics hochspezifische p27-Inhibitoren sein kann (kip1)-abh&auml;ngige VSMC Verbreitung nach vaskul&auml;re Sch&auml;digung.

Ma a differenza di rapamicina, cicaprost non inibire endoteliali nelle cellule endoteliali dell&#39;aorta anche quando utilizzato a concentrazioni &amp;gt; 10 volte superiore al suo ED(50) per VSMCs. Allo stesso modo, sia rapamicina e cicaprost sono stati segnalati per regolare i livelli dell&#39;inibitore cdk, p27(kip1). Ma rapamicina &egrave; rimasto anti-mitogenic in p27 (kip1)-null VSMCs considerando che l&#39;effetto anti-mitogenic di cicaprost era completamente dipendente da p27(kip1). Concludiamo che stabile PGI(2) mimetico pu&ograve; essere altamente specifici inibitori di p27 (kip1)-dipendente proliferazione VSMC dopo lesioni vascolari.

Los stents liberadores de f&#xE1;rmacos antiproliferativos limitar la reestenosis, pero las drogas de uso com&#xFA;n hasta la fecha son relativamente no espec&#xED;ficos agentes citost&#xE1;ticos que inhiben la proliferaci&#xF3;n de la &#xED;ntima c&#xE9;lulas endoteliales, as&#xED; como medial c&#xE9;lulas del m&#xFA;sculo liso y por lo tanto puede contribuir a las complicaciones cl&#xED;nicas asociadas a la angioplastia. En un esfuerzo para identificar una m&#xE1;s espec&#xED;fica anti-proliferativa agente, se compararon los efectos de la rapamicina a los de cicaprost, un mim&#xE9;tico de la natural anti-mit&#xF3;geno, IGP (2). La rapamicina y cicaprost fueron fuertemente anti-mitog&#xE9;nica en c&#xE9;lulas musculares lisas vasculares (CMLV). Pero a diferencia de la rapamicina, cicaprost no inhibe la mitog&#xE9;nesis en las c&#xE9;lulas endoteliales a&#xF3;rticas, incluso cuando se usa en concentraciones&#x26;gt; 10 veces por encima de su disfunci&#xF3;n er&#xE9;ctil (50) de CMLV. De manera similar, tanto la rapamicina y cicaprost han descrito para regular los niveles del inhibidor de CDK, p27 (kip1). Sin embargo, la rapamicina se mantuvo en la lucha contra el mit&#xF3;geno p27 (kip1) son nulos CMLV mientras que el efecto anti-mitog&#xE9;nica de cicaprost era completamente dependiente de p27 (kip1).
