Higher-order chromosomal organization for transcription regulation is poorly understood in eukaryotes. Using genome-wide Chromatin Interaction Analysis with Paired-End-Tag sequencing (ChIA-PET), we mapped long-range chromatin interactions associated with RNA polymerase II in human cells and uncovered widespread promoter-centered intragenic, extragenic, and intergenic interactions. These interactions further aggregated into higher-order clusters, wherein proximal and distal genes were engaged through promoter-promoter interactions. Most genes with promoter-promoter interactions were active and transcribed cooperatively, and some interacting promoters could influence each other implying combinatorial complexity of transcriptional controls. Comparative analyses of different cell lines showed that cell-specific chromatin interactions could provide structural frameworks for cell-specific transcription, and suggested significant enrichment of enhancer-promoter interactions for cell-specific functions. Furthermore, genetically-identified disease-associated noncoding elements were found to be spatially engaged with corresponding genes through long-range interactions. Overall, our study provides insights into transcription regulation by three-dimensional chromatin interactions for both housekeeping and cell-specific genes in human cells.

D&#x26;#39;ordre sup&#xE9;rieur pour l&#x26;#39;organisation chromosomique r&#xE9;gulation de la transcription est mal comprise chez les eucaryotes. En utilisant l&#x26;#39;&#xE9;chelle du g&#xE9;nome analyse de l&#x26;#39;interaction avec la chromatine appari&#xE9;s balise de fin de s&#xE9;quen&#xE7;age (Chia-PET), nous avons cartographi&#xE9; les interactions &#xE0; longue port&#xE9;e chromatine associ&#xE9;s &#xE0; l&#x26;#39;ARN polym&#xE9;rase II dans les cellules humaines et d&#xE9;couvert r&#xE9;pandues interactions promoteur-centr&#xE9;e intrag&#xE9;niques, extrag&#xE9;nique, et interg&#xE9;nique. Mit genomweiten Chromatin Interaktionsanalyse mit Paired-End-Tag-Sequenzierung (ChIA-PET), wir zugeordnet Langstrecken Chromatin-Interaktionen, die RNS-Polymerase II in menschlichen Zellen und ungedeckten weit verbreitete Projekttr&auml;ger-zentrierte intragenic, extragenic und intergenetischer Interaktionen zugeordnet. Diese Interaktionen aggregiert weiter in h&ouml;herer Ordnung Cluster aufgeteilt, wobei proximalen und distalen Gene durch Projekttr&auml;ger-Projekttr&auml;ger Interaktionen involviert waren. Die meisten Gene mit Projekttr&auml;ger-Projekttr&auml;ger Interaktionen waren aktiv und transkribierte kooperativ, und einige interagierende F&ouml;rderer konnte beeinflussen sich gegenseitig was bedeutet kombinatorischen Komplexit&auml;t transkriptionelle Steuerelemente. Vergleichende Analysen der verschiedenen Zelllinien zeigte, dass zellspezifische Chromatin Interaktionen k&ouml;nnte die strukturelle Rahmenbedingungen f&uuml;r zellspezifische Transkription bieten erhebliche Bereicherung der Vergr&ouml;&szlig;erer-Projekttr&auml;ger Interaktionen f&uuml;r zellspezifische Funktionen vorgeschlagen. Ordine superiore organizzazione cromosomica per regolazione trascrizione mal inteso negli eucarioti. Utilizzando l&#39;analisi di interazione della cromatina genome-wide con sequenziamento associati-fine-Tag (ChIA-PET), abbiamo mappato le interazioni a lungo raggio della cromatina associate con la RNA polimerasi II in cellule umane e scoperte diffuse promotore-centrato intragenic, extragenic e intergenic interazioni. Queste interazioni ulteriormente aggregate in ammassi di ordine superiore, in cui geni prossimali e distali sono stati impegnati attraverso interazioni Promotore-Organizzatore. La maggior parte dei geni con interazioni promoter-promotore erano attivi e trascritto in modo cooperativo, e alcuni promotori interagenti potrebbero influenzare l&#39;altro implicando complessit&agrave; combinatoria di transcriptional controlli. Analisi comparative di diverse variet&agrave; di cellula ha mostrato interazioni cellula-specifico della cromatina potrebbero fornire quadri strutturali per la trascrizione delle cellule specifiche che ha suggerito significativo arricchimento delle interazioni enhancer-promotore per funzioni specifiche di cella. Inoltre, identificati geneticamente associata a malattia non codificante elementi sono stati trovati per essere spazialmente impegnati con i geni corrispondenti attraverso interazioni a lungo raggio. Nel complesso, il nostro studio fornisce intuizioni nella regolazione della trascrizione di interazioni di cromatina tridimensionale per pulizia e cella specifici geni nelle cellule umane.

De orden superior la organizaci&#xF3;n cromos&#xF3;mica de regulaci&#xF3;n de la transcripci&#xF3;n es poco conocida en eucariotas. Uso de todo el genoma An&#xE1;lisis de Interacci&#xF3;n de la cromatina con la secuenciaci&#xF3;n pareadas etiqueta final (Chia-PET), que asigna a largo plazo las interacciones asociadas a la cromatina con la RNA polimerasa II en las c&#xE9;lulas humanas y descubrieron amplias interacciones promotor centrado en intrag&#xE9;nicos, extrag&#xE9;nicas y interg&#xE9;nica. Estas interacciones m&#xE1;s agregados en un grupo de orden superior, en el que los genes proximal y distal se dedicaban a trav&#xE9;s de la interacci&#xF3;n del promotor promotor. La mayor&#xED;a de los genes con las interacciones promotor promotor-se mantuvieron activos y transcrita en forma cooperativa, y algunos promotores que interact&#xFA;an entre s&#xED; puede influir en lo que implica la complejidad combinatoria de los controles de la transcripci&#xF3;n. An&#xE1;lisis comparativos de diferentes l&#xED;neas celulares mostraron que las c&#xE9;lulas espec&#xED;ficas de la cromatina interacciones podr&#xED;an proporcionar marcos estructurales de c&#xE9;lulas espec&#xED;ficas de la transcripci&#xF3;n, y sugiri&#xF3; un importante enriquecimiento de las interacciones potenciador-promotor de c&#xE9;lulas espec&#xED;ficas de las funciones. Por otra parte, gen&#xE9;ticamente determinadas enfermedades asociadas a los elementos no codificantes se comprob&#xF3; que estaban espacialmente comprometida con los genes correspondientes a trav&#xE9;s de interacciones de largo alcance. En general, nuestro estudio proporciona informaci&#xF3;n detallada sobre regulaci&#xF3;n de la transcripci&#xF3;n de la cromatina por las interacciones en tres dimensiones tanto para la limpieza y los genes espec&#xED;ficos de la c&#xE9;lula en las c&#xE9;lulas humanas.
