Een microchannels-on-a-chip platform werd ontwikkeld door de combinatie van fotolithografische reflowable fotolak techniek, zachte lithografie, en microfluidics. Het endotheel microkanalen platform bootst de driedimensionale (3D) geometrie van in vivo micro-vaatjes, loopt onder gecontroleerde continue perfusie stroom, zorgt voor hoge kwaliteit en real-time beeldvorming en kan worden toegepast voor microvasculaire onderzoek.
Inspanningen gericht op het ontwikkelen van in vitro assays voor de studie van microvaatjes omdat in vivo dierstudies zijn tijdrovend, duur en observatie en kwantificering zijn zeer uitdagend. Echter, conventionele in vitro assays microvaatje hebben beperkingen als die in vivo microvaatjes met betrekking tot driedimensionale (3D) geometrie en een continue vloeistofstroom. Met behulp van een combinatie van fotolithografische reflowable fotolak techniek, zachte lithografie, en microfluidics, hebben we een multi-diepte cirkelvormige dwarsdoorsnede endotheel microkanalen-on-a-chip, die de 3D-geometrie van de in vivo micro-vaatjes nabootst en draait onder gecontroleerde continue perfusie flow. Een positieve herplaatsbaar fotolak werd gebruikt om een meester mal te fabriceren met een halfronde dwarsdoorsnede microchannel netwerk. Door de afstemming en binding van de twee polydimethylsiloxaan (PDMS) microchannels replvoor bestemde van de meester mal, werd een cilindrische microchannel netwerk gecreëerd. De diameters van de microkanalen kan goed worden gecontroleerd. Bovendien primaire humane navelstreng endotheelcellen (HUVEC) gezaaid in de chip bleek dat de cellen gevoerd het binnenoppervlak van de microkanalen in gecontroleerde perfusie die gedurende een tijdsperiode tussen 4 dagen tot 2 weken.
Microvaatjes als onderdeel van het circulatiesysteem, bewerkstelligen de interactie tussen bloed en weefsels, ondersteunen metabolische activiteiten definiëren weefsel micro en spelen een belangrijke rol in vele gezondheids en pathologische omstandigheden. Recapitulatie van functionele microvaatjes in vitro kan een platform voor de studie van complexe vasculaire fenomenen. Echter, conventionele in vitro assays microvaatje, zoals endotheliale celmigratie testen buisvorming endotheliale assays en ratten en muizen aortaring assays, in staat zijn de opnieuw in vivo microvaatjes met betrekking tot driedimensionale (3D) geometrie als continu control 1-8. Studies microvaatjes met dierlijke modellen en in vivo assays, zoals corneale angiogenese assay, kuiken chorioallantoïsmembraan angiogenese assay en Matrigel plug assay, zijn tijdrovend, in hoge kosten, uitdaging met betrekking tot waarneming en kwantificering enethische kwesties 1, 9-13.
Vooruitgang in micromanufacturing en microfluïdische chip technologieën hebben verschillende inzichten mogelijk in biomedische wetenschappen terwijl vermindering de experimentele hoge kosten en complexiteit van de dieren in vivo studies 14, zoals gewoon strak gecontroleerde biologische omstandigheden en dynamische vloeibare milieus, die niet zou mogelijk met conventionele technieken macroschaal.
Hier presenteren we een aanpak om een endotheel construeren microchannels-on-a-chip, die de 3D-geometrie van de in vivo micro-vaatjes nabootst en draait onder gecontroleerde continue perfusie stroom door gebruik te maken van de combinatie van fotolithografische reflowable fotolak techniek, zachte lithografie, en microfluidics.
