The goal of this protocol is to use laser-capture micro-dissection as an effective method to isolate pure populations of cell types from heterogeneous prostate tissues for downstream RNA analysis.
The prostate gland contains a heterogeneous milieu of stromal, epithelial, neuroendocrine and immune cell types. Healthy prostate is comprised of fibromuscular stroma surrounding discrete epithelial-lined secretory lumens and a very small population of immune and neuroendocrine cells. In contrast, areas of prostate cancer have increased dysplastic luminal epithelium with greatly reduced or absent stromal population. Given the profound difference between stromal and epithelial cell types, it is imperative to separate the cell types for any type of downstream molecular analysis. Despite this knowledge, the bulk of gene expression studies compare benign prostate to cancer without micro-dissection, leading to stromal bias in the benign samples. Laser-capture micro-dissection (LCM) is an effective method to physically separate different cell types from a specimen section. The goal of this protocol is to show that RNA can be successfully isolated from LCM-collected human prostatic epithelium and used for downstream gene expression studies such as RT-qPCR and RNAseq.
De prostaat is een heterogeen weefsel samengesteld uit secretoire epitheel gerangschikt in glandular acini omgeven door fibromusculaire stroma voornamelijk samengesteld uit gladde spieren 1. De epitheliale compartiment omvat vijf verschillende maar georganiseerde celtypes: basale cellen, secretorische cellen, neuro-endocriene cellen, transit versterken en stamcellen 2. Bij prostaatkanker (PCa), die voortvloeit uit de luminale epitheliale cellen, de groei van het adenocarcinoom veroorzaakt een duidelijke geleidelijke afname van het stromale compartiment 3. Daarom zullen weefselmonsters zijn duidelijke verschillen in het aandeel van stromale en epitheliale celtype gebaseerd op de mate van PCa. Deze verschillen kunnen leiden tot vertekende aannames van de genexpressie gegevens van hele weefsels verkregen ongeacht microdissectie van het gewenste celtype. Daarom, om deze bias te verwijderen is het noodzakelijk om celtypen te scheiden voorafgaand aan RNA-extractie engenexpressie analyse.
Macrodissection of microdissectie kan worden fysiek gescheiden goed gekarakteriseerde epitheliale delen van de omliggende stroma 4 -6. Macrodissection wordt meestal gedaan met een scheermesje onder een dissectiemicroscoop en werkt goed voor het scheiden van grote prostaatkanker knobbeltjes stroma, maar niet kan verwijderen goedaardige epitheel van omliggende stroma (zie voorbeeld goedaardige prostaat histologie in figuur 1). Microdissectie met een laser (LCM) aanzienlijk arbeidsintensiever dan macrodissection, maar kan zeer nauwkeurig ontleden goedaardige epitheel 4.
Recente publicaties van ons lab hebben aangetoond dat RNA met succes door de LCM van beide formaline gefixeerde in paraffine ingebedde (FFPE) biopten of bevroren weefsel 4,7-9 kan worden gewonnen. De belangrijkste uitdagingen in LCM-RNA-extractie zijn 1) om nauwkeurig ontleden de gewenste gebieden van het weefsel, en 2) te behouden RNA integriteit tijdens de LCM en isolatie proces 4,10. RNA geïsoleerd uit zuivere celpopulaties kunnen worden gebruikt voor genexpressieanalyse door verscheidene werkwijzen zoals reverse transcriptie kwantitatieve PCR (RT-qPCR) 7,8, microarray 11 en diep -sequencing 12-14.
Het doel van dit protocol is het totaal RNA van LCM prostaat epitheel isoleren uit bevroren weefsel op stroomafwaartse genexpressie analyses.
Genexpressie profilering van humane monsters kan lastig zijn, niet alleen voor de kwaliteit of kwantiteit van weefsel mogelijk, maar ook de verschillende zich in een bepaald weefsel specimen histologische entiteiten. Deze Vooral in de prostaat waarin goedaardige weefsels grotendeels stromale weefsels en delen van kanker zijn verstoken van stroma.
