The in vivo measurement of smooth muscle contractions along the gastrointestinal tract of laboratory animals remains a powerful, though underutilized, technique. Flexible, dual element strain gages are not commercially available and require fabrication. This protocol describes the construction of reliable, inexpensive strain gages for acute or chronic implantation in rodents.
Gastrointestinal dysfunction remains a major cause of morbidity and mortality. Indeed, gastrointestinal (GI) motility in health and disease remains an area of productive research with over 1,400 published animal studies in just the last 5 years. Numerous techniques have been developed for quantifying smooth muscle activity of the stomach, small intestine, and colon. In vitro and ex vivo techniques offer powerful tools for mechanistic studies of GI function, but outside the context of the integrated systems inherent to an intact organism. Typically, measuring in vivo smooth muscle contractions of the stomach has involved an anesthetized preparation coupled with the introduction of a surgically placed pressure sensor, a static pressure load such as a mildly inflated balloon or by distending the stomach with fluid under barostatically-controlled feedback. Yet many of these approaches present unique disadvantages regarding both the interpretation of results as well as applicability for in vivo use in conscious experimental animal models. The use of dual element strain gages that have been affixed to the serosal surface of the GI tract has offered numerous experimental advantages, which may continue to outweigh the disadvantages. Since these gages are not commercially available, this video presentation provides a detailed, step-by-step guide to the fabrication of the current design of these gages. The strain gage described in this protocol is a design for recording gastric motility in rats. This design has been modified for recording smooth muscle activity along the entire GI tract and requires only subtle variation in the overall fabrication. Representative data from the entire GI tract are included as well as discussion of analysis methods, data interpretation and presentation.
Experimentele studies dat record in vivo gastro-intestinale (GI) motiliteit over een aantal experimentele condities blijft een krachtig hulpmiddel voor het begrijpen van de onderliggende normale en pathofysiologische processen die nodig zijn voor de voedingsstof homeostase. Traditioneel talrijke experimentele methoden, sommige met overeenkomsten met die in de klinische praktijk 1, zijn gebruikt om direct veranderingen in GI contractie rate 2-5, intraluminale druk 6, 7, of GI doorvoer van niet-absorbeerbare markers 8, 9 kwantificeren of stabiele isotopen 10-12. Elk van deze technieken heeft unieke voordelen en nadelen die eerder zijn besproken in de literatuur. Zo is het nut van de ballon manometrie op drukverschillen kwantificeren ondervraagd vanwege de inherente naleving van de ballon materiaal terwijl maag-herstel van niet-absorbeerbare markers vereist euthanizing de experimentele animal voor een enkel datapunt. Onlangs heeft de toepassing en validering van een geminiaturiseerde arteriële druk catheter gerapporteerd dat een niet-chirurgische methode voor het monitoren maag contractiliteit in ratten en muizen 3 biedt. Terwijl een orogastrically geplaatst drukomzetter elimineert effectief verstorende variabelen gastrointestinale functie door het vermijden invasieve chirurgische procedures, dergelijke benadering alleen voor verdoofd preparaten. Bovendien heeft het ontbreken van visuele geleiding niet consistent plaatsing van de transducer binnen bepaalde gebieden van de maag mogelijk. Als zodanig is deze toepassing beperkt tot de maag of darm sinds visualisatie, in combinatie met de relatief stijve draad transducer binnen de duodenum en ileum geen optie.
Ook heeft de biomagnetisch wisselstroom biosusceptometry (ACB) techniek gevalideerd voor GI krimp analyse 4. Terwijl de ACB-techniek biedt een niet-invasieve aptoeloopt voor het meten maag contracties, ACB lijdt een soortgelijke beperking in het gebruik van opgenomen magnetische detectie media ofwel geen nauwkeurige registratie van specifieke gebieden van het maagdarmkanaal mogelijk. Deze beperking kan worden overwonnen door chirurgische implantatie van magnetische markers. Toch is de ACB techniek vereist dat de dieren verdoofd worden voor het verzamelen van gegevens.
Ultrasonomicrometry is werkzaam in een aantal GI studeert 13, 14 om te profiteren van de geringe omvang, ruimtelijke en temporele voordelen van piëzo-elektrisch kristal zender / ontvangers. Golven van maag gladde spiercontractie geen hoogfrequente event en optreden met een snelheid van ongeveer 3-5 cycli / min. Daarom kan de temporele voordelen van sonomicrometry behoeven de kosten te rechtvaardigen. Bovendien, terwijl de lineaire beweging nauwkeurig wordt gemeten met sonomicrometry, beperkingen werden ingediend met betrekking accurate maag-gegevensinterpretatie die kan voortvloeien uit het implanteren van een onvoldoende aantal kristallen 14.
Op basis van de originele designs Bass en collega 2 15 Dit protocol gevisualiseerd vollediger worden de stap-voor-stap fabricage en experimentele toepassing van mini, twee elementen rekstrookjes die hoge gevoeligheid en flexibiliteit voor de registratie gladde spiercontracties langs de gehele GI tract. De afmetingen van de rekstrook elementen zijn geschikt voor elke knaagdier was sinds gevoeligheid en de afgewerkte rekstrookje meest afhankelijk van de silicone sheets inkapselen van de elementen. Deze rekstrookjes worden gemakkelijk aangepast voor acute en chronische toepassing in verdoofde en vrij gedragen proefdiermodellen waardoor een enkele techniek voor het kwantificeren van gladde spiercontracties.
De hier gepresenteerde procedures kan elk laboratorium gevoelige miniatuur rekstrookjes fabriceren voor biologische toepassingen waaronder, maar niet beperkt tot, gastro-intestinale motiliteit in kleine proefdieren. Omdat de commerciële vervaardiging van deze rekstrookjes opgehouden worden laboratoria onderzoeken maagdarmfunctie beperkt tot andere technieken die de reeks experimentele toepassingen die beschikbaar zijn kunnen toestaan.
