We present a method for microfluidic deposition of patterned genipin and fibronectin on PDMS substrates, allowing extended viability of vascular smooth muscle cell-dense tissues. This tissue fabrication method is combined with previous vascular muscular thin film technology to measure vascular contractility over disease-relevant time courses.
The chronic nature of vascular disease progression requires the development of experimental techniques that simulate physiologic and pathologic vascular behaviors on disease-relevant time scales. Previously, microcontact printing has been used to fabricate two-dimensional functional arterial mimics through patterning of extracellular matrix protein as guidance cues for tissue organization. Vascular muscular thin films utilized these mimics to assess functional contractility. However, the microcontact printing fabrication technique used typically incorporates hydrophobic PDMS substrates. As the tissue turns over the underlying extracellular matrix, new proteins must undergo a conformational change or denaturing in order to expose hydrophobic amino acid residues to the hydrophobic PDMS surfaces for attachment, resulting in altered matrix protein bioactivity, delamination, and death of the tissues.
Here, we present a microfluidic deposition technique for patterning of the crosslinker compound genipin. Genipin serves as an intermediary between patterned tissues and PDMS substrates, allowing cells to deposit newly-synthesized extracellular matrix protein onto a more hydrophilic surface and remain attached to the PDMS substrates. We also show that extracellular matrix proteins can be patterned directly onto deposited genipin, allowing dictation of engineered tissue structure. Tissues fabricated with this technique show high fidelity in both structural alignment and contractile function of vascular smooth muscle tissue in a vascular muscular thin film model. This technique can be extended using other cell types and provides the framework for future study of chronic tissue- and organ-level functionality.
Vasculaire ziekten, zoals cerebrale vasospasmen 1,2, 3 hypertensie en atherosclerose 4, langzaam ontwikkelen, zijn gewoonlijk chronisch van aard en omvatten disfunctionele kracht generatie door vasculaire gladde spiercellen (VSMC's). Wij streven ernaar om deze langzaam vordert vasculaire disfuncties met in vitro methoden met fijnere controle van de experimentele omstandigheden dan in in vivo modellen te bestuderen. We hebben eerder ontwikkelde vasculaire gespierde dunne films (vMTFs) voor het meten van functionele contractiliteit van in vitro gemanipuleerde cardiovasculaire weefsels 5, maar deze methode is beperkt tot een relatief korte termijn studies. Hier presenteren we een substraat modificatie techniek die onze vorige vMTF techniek breidt voor langdurige metingen.
Terwijl het endotheel is ook kritisch in algemene vasculaire functie ontworpen arteriële lamellen een bruikbaar model systeem voor het vastleggen van veranderingen in vasculairecontractiliteit tijdens progressie van de ziekte. Een functionele vaatziekte weefselmodel zowel de structuur en functie van de arteriële lamel ingenieur, de basis contractiele apparaat van het vaartuig moet worden recapituleerde een hoge getrouwheid. Arteriële lamellen concentrisch omtrek uitgelijnde vellen contractiele VSMC gescheiden vellen van elastine 6. Microcontact printen van extracellulaire matrix (ECM) eiwitten op polydimethylsiloxaan (PDMS) substraten is eerder gebruikt als leidraad signalen voor weefselorganisatie nabootsen uitgelijnd cardiovasculair weefsel 5,7-10. Echter, weefsels gevormd via microcontact afdrukken kan integriteit verliezen na 3-4 dagen kweken beperking van hun toepasbaarheid bij chronische studies. Dit protocol voorziet in een oplossing voor dit probleem door het vervangen van de vorige microcontact druktechnieken met een nieuwe microfluïdische depositie techniek.
Genchi et al. Gemodificeerde PDMS substraten met genipin en found langdurige levensvatbaarheid van myocytes tot een maand in de cultuur 11. Hier gebruiken we een soortgelijke benadering van cultuur van patroon vasculaire gladde spiercellen te breiden op PDMS. Genipin, natuurlijke hydrolytische afgeleide van de gardenia fruit, een gewenste kandidaat voor modificatie substraat vanwege de relatief lage toxiciteit in vergelijking met soortgelijke verknopingsmiddelen en het toenemende gebruik als een biomateriaal in het gebied van weefselherstel 12,13 en ECM modificatie 14, 15. In dit protocol wordt fibronectine gebruikt als een cel begeleiding cue, net als in voorgaande microcontactprinten methoden; echter genipin afgezet op PDMS substraten voorafgaand aan fibronectine patronen. Dus, zoals cellen afbreken van de matrix patroon, nieuw gesynthetiseerde ECM van aangesloten VSMC kan binden aan het PDMS-genipin beklede substraat.
Dit protocol maakt gebruik van een microfluïdische afleverinrichting voor tweefasige genipin en ECM afzetting. Het ontwerp van de microfluïdische apparaat bootst microcontact drukpatronen ontworpen voor arteriële lamellen in eerdere studies 16. Zo verwachten we dat dit protocol om arteriële lamellen bootst dat succes herhalen de zeer uitgelijnd in vivo structuur en de contractiele functie van de arteriële lamellen opleveren. We hebben ook weefsel contractiliteit te evalueren om te bevestigen dat genipin een geschikt substraat modificatie compound voor langdurige in vitro vasculaire ziektemodellen.
Hier presenteren we een protocol dat gebaseerd is op eerder ontwikkelde vMTF technologie, waardoor langere experiment keer meer kenmerkend vasculaire chronische ziekteprocessen 1,23,24. Hiervoor micropatroon we genipin, die vooraf is aangetoond dat langdurige functionalisering van PDMS substraten 11 indienen waarbij een microfluidic depositietechniek op gemanipuleerde arteriële lamellen met verbeterde vasculair levensvatbaarheid van weefsel voor gebruik in MTF contractiliteitsstudies verkregen.
