Axon guidance molecules regulate neuronal migration and targeted growth-cone navigation. We present a powerful method, the stripe assay, to assess the ability of guidance molecules to attract or repulse neurons. In this protocol, we demonstrate the stripe assay by showing FLRT2’s ability to repel cultured hippocampal neurons.
Growing axons develop a highly motile structure at their tip, termed the growth cone. The growth cone contacts extracellular environmental cues to navigate axonal growth. Netrin, slit, semaphorin, and ephrins are known guidance molecules that can attract or repel axons upon binding to receptors and co-receptors on the axon. The activated receptors initiate various signaling molecules in the growth cone that alter the structure and movement of the neuron. Here, we describe the detailed protocol for a stripe assay to assess the ability of a guidance molecule to attract or repel neurons. In this method, dissociated hippocampal neurons from E15.5 mice are cultured on laminin-coated dishes processed with alternating stripes of ectodomain of fibronectin and leucine-rich transmembrane protein-2 (FLRT2) and control immunoglobulin G (IgG) fragment crystallizable region (Fc) protein. Both axons and cell bodies were strongly repelled from the FLRT2-coated stripe regions after 24 h of culture. Immunostaining with tau1 showed that ~90% of the neurons were distributed on the Fc-coated stripes compared to the FLRT2-Fc-coated stripes (~10%). This result indicates that FLRT2 has a strong repulsive effect on these neurons. This powerful method is applicable not only for primary cultured neurons but also for a variety of other cells, such as neuroblasts.
Axon begeleiding is het proces waarbij nieuw gevormde neuronen sturen hun axonen Tijdens het ontwikkelen van het zenuwstelsel 1,2. Het ontwikkelen van axonen dragen een zeer beweeglijke structuur op hun tip genaamd de groei kegel. De groei kegel zintuigen extracellulaire signalen naar het pad van de axon te navigeren. Navigatie moleculen, zoals spleet semaphorines en ephrines kunnen aantrekken of afstoten axons afhankelijk van hun wisselwerking met geschikte receptoren en co-receptoren op het axon 1,3,4. De geactiveerde receptoren dragen signalen naar de groeikegel dat de organisatie van het cytoskelet en van axon groei-cone bewegingen beïnvloeden.
Verschillende werkwijzen zijn ontwikkeld om de werking van aantrekkende en afstotende moleculen evalueren. Chemo-aantrekkende en insectenwerende middelen kunnen worden toegediend in de groei / cultuurmedium met een gradiënt concentratie (bv., Dunn's kamer of μ-dia) 5,6, in een zeer geconcentreerde spot door micro-pipette (bijv., draaien bepaling) 7 of op een homogene concentratie bad toepassing (bv., groeikegel ineenstorting test) 8,9.
Andere werkwijzen omvatten een streep assay of microcontactprinten (μCP), waarin een chemo-aantrekkende of afstotende wordt gecoat op het oppervlak van een plaat als substraat 10-12. Thestripe test werd oorspronkelijk ontwikkeld door Bonhoeffer en collega's in 1987 tot topografische kaart brengen in het kuiken retino-tectal systeem 13 te analyseren. De oorspronkelijke methode vereist een vacuüm systeem om manteleiwitten op polycarbonaat nucleopore membranen met behulp van gestreepte en fijnmazige matrices. In latere versies werden de recombinante eiwitten direct gedrukt op het oppervlak van een kweekplaat in een gestreept patroon via smalle spleet silicium matrices 14,15. Onlangs hebben verschillende onderzoeksgroepen succes toegepast deze streep test om de analyse van axon begeleiding moleculen activiteiten 16-21.
<p class = "jove_content"> Hier presenteren we het gedetailleerd protocol voor een streep test die de aantrekking of afstoting van axon begeleiding moleculen voor gedissocieerde hippocampus neuronen meet. Met name kan deze werkwijze worden toegepast bij minimaal uitgerust laboratorium instellingen. Voor deze test zijn afwisselende strepen van een fluorescent gelabeld substraat en een controle-eiwit gegenereerd op een plastic schaaltje met een siliconen matrix met 90-um spleten en gecoat met laminine. In onze demonstratie, los hippocampus neuronen uit E15.5 muizen werden gekweekt op afwisselende strepen van recombinante ectodomein van fibronectine en leucine-rijke transmembraaneiwit-2 (FLRT2) en de controle Fc proteïne 21. Na 24 uur kweken werden zowel de axonen en cellichamen van de neuronen sterk afgestoten van de FLRT2 strepen. Kleuring met anti-Tau1 antilichaam bleek dat ~ 90% van de neuronen werden verspreid op-Fc beklede gebieden, tegenover ~ 10% op de FLRT2-Fc, wat aangeeft dat FLRT2 een sterke afstotendefunctie hippocampale neuronen 21.Dit protocol beschrijft een streep test die recombinant eiwit en gedissocieerde neuronen maakt gebruik van het E15.5 muis hippocampus. Deze test maakt het mogelijk efficiënte observatie van afstotelijk, aantrekkelijk, of neutraal reacties van neuronen om een recombinant eiwit van interesse in een gestreept patroon.
