Hier wordt een protocol gepresenteerd voor de bereiding en bufferkalibratie van celextracten uit exonuclease V knock-out stammen van Escherichia coli BL21 Rosetta2 (ΔrecBCD en ΔrecB). Dit is een snelle, eenvoudige en directe benadering voor expressie in celvrije eiwitsynthesesystemen met behulp van lineaire DNA-sjablonen.
