The use of fluorophores for in vivo imaging can be greatly limited by opsonization, rapid clearance, low detection sensitivity and cytotoxic effects on the host. Encapsulation of fluorophores in liposomes by film hydration and extrusion leads to fluorescence quenching and protection which enables in vivo imaging with high detection sensitivity.
Optical imaging offers a wide range of diagnostic modalities and has attracted a lot of interest as a tool for biomedical imaging. Despite the enormous number of imaging techniques currently available and the progress in instrumentation, there is still a need for highly sensitive probes that are suitable for in vivo imaging. One typical problem of available preclinical fluorescent probes is their rapid clearance in vivo, which reduces their imaging sensitivity. To circumvent rapid clearance, increase number of dye molecules at the target site, and thereby reduce background autofluorescence, encapsulation of the near-infrared fluorescent dye, DY-676-COOH in liposomes and verification of its potential for in vivo imaging of inflammation was done. DY-676 is known for its ability to self-quench at high concentrations. We first determined the concentration suitable for self-quenching, and then encapsulated this quenching concentration into the aqueous interior of PEGylated liposomes. To substantiate the quenching and activation potential of the liposomes we use a harsh freezing method which leads to damage of liposomal membranes without affecting the encapsulated dye. The liposomes characterized by a high level of fluorescence quenching were termed Lip-Q. We show by experiments with different cell lines that uptake of Lip-Q is predominantly by phagocytosis which in turn enabled the characterization of its potential as a tool for in vivo imaging of inflammation in mice models. Furthermore, we use a zymosan-induced edema model in mice to substantiate the potential of Lip-Q in optical imaging of inflammation in vivo. Considering possible uptake due to inflammation-induced enhanced permeability and retention (EPR) effect, an always-on liposome formulation with low, non-quenched concentration of DY-676-COOH (termed Lip-dQ) and the free DY-676-COOH were compared with Lip-Q in animal trials.
Liposomen zijn intensief onderzocht en dienen als een van de meest biocompatibele biomedische geneesmiddelafgiftesystemen voor klinische toepassingen 1,2. Zij bestaan hoofdzakelijk uit fosfolipiden en cholesterol, die beide biocompatibele verbindingen nabootsen delen van natuurlijke celmembranen. Overwegende dat hydrofiele stoffen kunnen worden ingesloten in de waterige interieur, kan lipofiele agenten binnen het liposomaal fosfolipidedubbellaag 3 worden opgenomen. Inkapseling van stoffen in het waterige inwendige van liposomen bescherming biedt tegen afbraak in vivo en voorkomt ook het hostsysteem van toxische effecten van chemotherapie voor de behandeling van ziekten, bijvoorbeeld chemotherapeutica gericht op het vernietigen van tumorcellen. De wijziging van de liposoomoppervlakte met polymeren zoals polyethyleenglycol (PEGylering) verdere uitbreiding van de liposomaal bloedcirculatie tijd in vivo vanwege sterische stabilisatie 4. Moreoveh, liposomen kunnen afzonderen hoge concentraties van verschillende stoffen zoals eiwitten 5,6, hydrofiele stoffen 7,8 en enzymen 9. Ze dienen dan ook als betrouwbare klinische therapeutische en diagnostische instrumenten die hun goedkeuring voor de levering van cytotoxische geneesmiddelen verdienen, zoals doxorubicine voor kankertherapie 4. Door hun flexibiliteit kunnen liposomen worden geladen met fluorochromen voor diagnostische en beeldgeleide chirurgische doeleinden.
Fluorescentiebeeldvorming levert een rendabele en niet-invasief in vivo diagnostisch hulpmiddel vereist echter een aantal fundamentele eisen. Het kan worden aangetoond dat fluorchromen die het best voor in vivo beeldvorming past hebben karakteristieke absorptie en emissie maxima in het bereik waar het licht dispersie en verstrooiing evenals weefsel autofluorescence afkomstig van water en hemoglobine is laag. Aldus dergelijke probes hun abs / em maxima tussen 650 en 900 nm 10. Daarnaast, de stabiliteit van de fluorochromen zowel in vitro pt in vivo is kritisch, aangezien opsonisatie en snelle klaring sterk kunnen beperken hun toepassing in vivo beeldvorming 11. Andere effecten, zoals slechte stabiliteit en een lage gevoeligheid of cytotoxische effecten op doelorganen zoals gezien met indocyaninegroen (ICG) 12-16, zijn ongewenst en moet in aanmerking worden genomen bij het ontwerpen van sondes voor in vivo beeldvorming. Deze waarnemingen hebben geleid tot de actieve ontwikkeling van verschillende preklinische NIR fluorochromen, nanodeeltjes en nieuwe technieken voor de in vivo beeldvorming van ontstekingsprocessen, kanker en voor beeldgeleide chirurgie 17-20. Ondanks de stabiliteit van de meeste preklinische NIRF (nabij-infrarood fluorescentie) kleurstoffen in vitro, hun snelle perfusie en klaring door de lever en nieren belemmeren het gebruik bij de in vivo optische beeldvorming van ziekten en ontstekingsprocessen.

Bij hoge concentraties H- dimeervorming en / of pi stapelen interacties tussen fluorofoor moleculen zich binnen elkaars Förster straal resultaat Förster resonantie energieoverdracht (FRET) tussen het fluorochroom moleculen. Bij lage concentraties de ruimte tussen de fluorofoor moleculen toeneemt, waardoor-pi stapelen wisselwerking voorkomen en H-dimeervorming en resulteert in een hoge fluorescentie-emissie. De schakelaar tussen hoge en lage concentratie en de bijbehorende fluorescentiedoving en activering is een veelbelovende strategie die kan worden benut voor optische beeldvorming 22. Hierbij inkapselen van hoge concentraties van de kleurstof NIRF DY-676-COOH in het waterige inwendige van liposomen is favorable voor in vivo beeldvorming dan de vrije kleurstof. De uitdaging van de werkwijze ligt allereerst in de juiste inkapseling en anderzijds in de validering van de voordelen van het inkapselen van hoge concentraties van de kleurstof. Vergelijking van de beeldvormende eigenschappen van liposomen geblust met die van de vrije kleurstof en ook een niet afgeschrikt liposoomformulering met lage concentraties van de kleurstof onontbeerlijk. We tonen door een eenvoudige maar zeer effectieve film hydratatie en extrusie protocol gecombineerd met afwisselend bevriezen en ontdooien cycli die inkapseling van uitdoving concentraties DY-676-COOH in liposomen haalbaar. Andere methoden voor het bereiden van liposomen zoals de omgekeerde fase verdamping werkwijze 23 en de ethanol injectiemethode 24 staat liposoompreparaat hoge efficiënties van inkapseling voor veel hydrofiele stoffen. De aard van de stof te kapselen kan de inkapseling efficiëntie beïnvloeden. In effect,de film hydratatie en extrusie-protocol hier gepresenteerde onthulde het hoogste rendement voor het inkapselen van DY-676-COOH. Om de voordelen van liposomale inkapseling van DY-676-COOH, een zymosan geïnduceerde oedeem model, dat de studie van ontstekingsprocessen binnen enkele uren laat illustreren, werd gebruikt. Hier wordt aangetoond dat liposomen met een hoge concentratie van het ingekapselde DY-676-COOH zijn meer geschikt voor het gehele lichaam in vivo optische beeldvorming van ontstekingsprocessen dan de vrije kleurstof of niet- afgeschrikt liposomale formulering met lage concentraties kleurstof. Dus de onderliggende protocol een eenvoudige en snelle methode om geblust fluorescerende liposomen en de validatie van de activering en imaging potentieel zowel in vitro als in vivo produceren.Aangezien liposomen ook kan dienen als afgiftesystemen voor fluorescente kleurstoffen, zij stellen beeldvorming van doelziekten. Het inkapselen van hoge concentraties van fluorescente kleurstoffen zoals NIRF kleurstof, DY676-COOH hier gebruikt, leidt tot een hoog niveau van fluorescentie uitdoving van de ingesloten kleurstof.
