The use of fluorophores for in vivo imaging can be greatly limited by opsonization, rapid clearance, low detection sensitivity and cytotoxic effects on the host. Encapsulation of fluorophores in liposomes by film hydration and extrusion leads to fluorescence quenching and protection which enables in vivo imaging with high detection sensitivity.
Optical imaging offers a wide range of diagnostic modalities and has attracted a lot of interest as a tool for biomedical imaging. Despite the enormous number of imaging techniques currently available and the progress in instrumentation, there is still a need for highly sensitive probes that are suitable for in vivo imaging. One typical problem of available preclinical fluorescent probes is their rapid clearance in vivo, which reduces their imaging sensitivity. To circumvent rapid clearance, increase number of dye molecules at the target site, and thereby reduce background autofluorescence, encapsulation of the near-infrared fluorescent dye, DY-676-COOH in liposomes and verification of its potential for in vivo imaging of inflammation was done. DY-676 is known for its ability to self-quench at high concentrations. We first determined the concentration suitable for self-quenching, and then encapsulated this quenching concentration into the aqueous interior of PEGylated liposomes. To substantiate the quenching and activation potential of the liposomes we use a harsh freezing method which leads to damage of liposomal membranes without affecting the encapsulated dye. The liposomes characterized by a high level of fluorescence quenching were termed Lip-Q. We show by experiments with different cell lines that uptake of Lip-Q is predominantly by phagocytosis which in turn enabled the characterization of its potential as a tool for in vivo imaging of inflammation in mice models. Furthermore, we use a zymosan-induced edema model in mice to substantiate the potential of Lip-Q in optical imaging of inflammation in vivo. Considering possible uptake due to inflammation-induced enhanced permeability and retention (EPR) effect, an always-on liposome formulation with low, non-quenched concentration of DY-676-COOH (termed Lip-dQ) and the free DY-676-COOH were compared with Lip-Q in animal trials.
Liposomer har blitt intensivt undersøkt og tjener som en av de biokompatible biomedisinske medikamentleveringssystemer for kliniske anvendelser 1,2. De er hovedsakelig sammensatt av fosfolipider og kolesterol, som begge er biokompatible forbindelser som etterligner deler av naturlige cellemembraner. Mens hydrofile stoffer kan bli innesluttet i det vandige indre, kan lipofile midler innarbeides i liposomalt fosfolipid dobbeltlag 3. Innkapsling av stoffer i det vandige indre av liposomer gir beskyttelse mot degradering in vivo, og hindrer også at vertssystemet fra toksiske effekter av cytotoksiske medikamenter anvendt for behandling av sykdommer, for eksempel kjemoterapeutika som tar sikte på å ødelegge tumorceller. Modifikasjonen av den liposomale overflaten med polymerer som polyetylenglykol (PEGylering) videre strekker den liposomale blodsirkulasjonstiden in vivo på grunn av sterisk stabilisering 4. Moreoveh, liposomer kan sekvestrere høye konsentrasjoner av flere stoffer slik som proteiner 5,6, hydrofile substanser og enzymer 7,8 9. De tjener derfor som pålitelige kliniske terapeutiske og diagnostiske verktøy som fortjener sin godkjenning for levering av cytotoksiske legemidler som doxorubicin for kreftbehandling 4. På grunn av sin fleksibilitet, kan liposomer også være lastet med fluorokromer for diagnostiske og bildestyrt kirurgi formål.
Fluorescens bildebehandling gir en kostnadseffektiv og ikke-invasiv in vivo diagnostisk verktøy som imidlertid krever noen grunnleggende krav. Det kunne påvises at fluorokromer som passer best for in vivo avbildning har karakteristisk absorpsjon og emisjon maksima i området hvor lysspredning og spredning, så vel som vev autofluorescens som stammer fra vann og hemoglobin er lav. Dermed slike sonder har sin abs / em maxima mellom 650 og 900 nm 10. I tillegg til dette, er stabiliteten av fluorokromer både in vitro og in vivo kritisk, som opsonisering og rask klaring kan i stor grad begrenser deres anvendelse for in vivo avbildning 11. Andre effekter som dårlig stabilitet og lav følsomhet eller cytotoksiske effekter på målorganer som sett med indocyanine grønn (ICG) 12-16, er uønsket og må tas hensyn til ved utformingen av sonder for in vivo imaging. Disse observasjonene har ført til aktiv utvikling av flere prekliniske NIR fluorokromer, nanopartikler samt nye teknikker for in vivo avbildning av inflammatoriske prosesser, kreft og for bildestyrt kirurgi 17-20. Til tross for stabiliteten av de prekliniske NIRF (nær infrarød fluorescens) fargestoffer som in vitro, er deres hurtige perfusjon og klaring gjennom leveren og nyrene hindrer deres anvendelse i in vivo optisk avbildning av sykdommer og inflammatoriske prosesser.
ntent "> Vi vil derfor presentere en protokoll for innkapsling av fluorokromer slik som godt karakterisert nær-infrarøde fluorescerende fargestoff DY-676-COOH som er kjent for sin tendens til å selv-avkjølings ved relativt høye konsentrasjoner 21 i liposomer. Ved høye konsentrasjoner H- dimer-dannelse og / eller pi-stabling interaksjoner mellom fluoroformolekyler som befinner seg innenfor hverandres Förster radius resultat Forster resonans energioverføring (FRET) mellom fluorokromkonjugerte molekyler. Ved lav konsentrasjon rommet mellom fluoroformolekyler øker, for derved å hindre pi-stabling interaksjon og H-dimer-dannelse og som resulterer i høy fluorescensemisjonen. Bryteren mellom høy og lav konsentrasjon, og det medfølgende fluorescensslukking og aktivisering er en lovende strategi som kan utnyttes for optisk avbildning 22. I dette henseende, innkapsling av høye konsentrasjoner av NIRF fargestoff DY-676-COOH i den vandige indre av liposomer er flere favorable for in vivo avbildning enn den gratis fargestoff. Utfordringen av metoden ligger først og fremst i riktig innkapsling og for det andre, i valideringen av fordelene som følger av innkapsle høye konsentrasjoner av fargestoffet. Sammenligning av de bildedannende egenskaper av bråtilsatt liposomer med det av den frie fargestoff og også med en ikke-undertrykket liposomformulering med lave konsentrasjoner av fargestoffet er uunnværlig. Vi viser av en enkel, men svært effektive film hydrering og ekstrudering protokoll kombinert med alternative fryse og tinesykluser som innkapsling av slukke konsentrasjoner av DY-676-COOH i liposomer er gjennomførbart. Andre metoder som brukes for å fremstille liposomer for eksempel revers fase fordamping metode 23 samt etanolinjeksjonsmetoden 24 aktiverer liposompreparat med høye innkapslingseffektiviteter for mange hydrofile stoffer. Imidlertid arten av den substans som skal innkapsles kan påvirke innkapslingseffektivitet. I praksisfilmen hydrering og ekstrudering protokollen presenteres her avslørte den høyeste effektivitet for innkapsling av DY-676-COOH. For å illustrere fordelene ved liposomal innekapsling av DY-676-COOH, en zymosan-induserte ødemmodell, som tillater undersøkelse av inflammatoriske prosesser innen noen få timer, ble anvendt. Her er det vist at liposomer med høye konsentrasjoner av den innkapslede DY-676-COOH er mer egnet for hele kroppen in vivo optisk avbildning av inflammatoriske prosesser enn det frie fargestoff eller den ikke-undertrykket liposomal formulering med lav fargestoffkonsentrasjoner. Således underliggende protokoll gir en enkel og rask metode for å produsere slukket fluorescerende liposomer og validering av deres aktivering og avbildning potensial både in vitro og in vivo.Siden liposomer kan også tjene som leveringssystemer for fluorescerende fargestoffer, aktiverer de avbildning av målet sykdommer. Innkapslingen av høye konsentrasjoner av fluorescerende fargestoffer som NIRF fargestoff, DY676-COOH anvendes her, fører til en høy grad av fluorescensslukking av innesluttet fargestoff.
