Environmental enrichment provides a potential protective effect against neurodegenerative disorders. Currently, however, there is no easy way of determining the efficacy of enrichment procedures. This protocol describes a simple “Puzzle Box” method for assessing an animal’s cognitive function, in order to reveal the effectiveness of environmental enrichment.
Environmental enrichment can dramatically influence the development and function of neural circuits. Further, enrichment has been shown to successfully delay the onset of symptoms in models of Huntington’s disease 1-4, suggesting environmental factors can evoke a neuroprotective effect against the progressive, cellular level damage observed in neurodegenerative disorders. The ways in which an animal can be environmentally enriched, however, can vary considerably. Further, there is no straightforward manner in which the effects of environmental enrichment can be assessed: most methods require either fairly complicated behavioral paradigms and/or postmortem anatomical/physiological analyses. This protocol describes the use of a simple and inexpensive behavioral assay, the Puzzle Box 5-7 as a robust means of determining the efficacy of increased social, sensory and motor stimulation on mice compared to cohorts raised in standard laboratory conditions. This simple problem solving task takes advantage of a rodent’s innate desire to avoid open enclosures by seeking shelter. Cognitive ability is assessed by adding increasingly complex impediments to the shelter’s entrance. The time a given subject takes to successfully remove the obstructions and enter the shelter serves as the primary metric for task performance. This method could provide a reliable means of rapidly assessing the efficacy of different enrichment protocols on cognitive function, thus paving the way for systematically determining the role specific environmental factors play in delaying the onset of neurodevelopmental and neurodegenerative disease.
Miljöberikning (EE) kan definieras som omgivningen som ger djur med ökad möjlighet till social interaktion, motorik och större sensorisk stimulering än vanligt upplevt i ett standardlaboratorie environment.EE har visat sig konsekvent påverkar djurens beteende, att få till stånd förändringar såsom minskad stress och ångest relaterad aktivitet 8-10, förbättrad prestanda i inlärnings- och minnesuppgifter 8,11, tidig debut av motor samordning och utforskande aktivitet 11, förändringar i mödravård 8 samt resistens mot beroendeframkallande ämnen 12- 15. Vidare har EE avslöjats att lindra effekterna av neurodegenerativa sjukdomar, fördröja debuten och minska svårighetsgraden av symtom i djurmodeller av Huntingtons 1-4,16, Parkinson 17 och Alzheimers sjukdom 18.
Dessa förändringar correlatera de anatomiska och molekylära förändringar EE är känt för att framkalla hela hjärnan. Djur som fötts upp i inredda miljöer från tidiga stadier av utveckling visar en myriad av neurala förändringar, bland annat ökad hjärnvikt och kortikal tjocklek, dendritiska förgrening 9,2-22 och synaptisk densitet 23. EE kan förändra både nivån och tidpunkten för tillväxtfaktor uttryck 9,24-30, vilket har visat sig bidra till snabbare utveckling av sensoriska 25,26,28,29, mnemonic 30, samt motorkretsar 31,32.
Tidigare arbete har avslöjat ibland motsägelse fynd vid undersökning av effekterna av EE, utan att ta hänsyn till de olika typer av djur och miljöer som används inom enskilda studier 9,24,27,30. För närvarande finns det ingen konsekvent och enkel beteende uppgift som kan användas för att mäta effektiviteten av olika EE paradigm i olika stammar och spectalet av djur.
Pusslet Box uppgift var utformad som ett enkelt test för att fastställa ett djurs infödda problemlösningsförmåga 7. Djur som placeras i det öppna området är skyldiga att ta bort hindrande material belägna inom en liten öppning för att få tillgång till en täckt region / kvinnojour. Varje ämne ges tre försök med samma hinder för att bedöma tre olika kognitiva attribut. Den första rättegången ger en baslinje indikation på inneboende eller infödda problemlösningsförmåga. Den andra rättegången, köras på samma dag, ger en fingervisning om djurets förmåga att förbättra och därmed förstärka strategier för att ta bort den specifika hindret. Den tredje studien, som genomfördes på följande dag, ger en inblick i förmåga motivet att behålla och återkalla lärde lösningen till uppgiften.
Motivationen för att lösa dessa "obstruktion pussel" av djuren kan varieras, potentielltmana en medfödd önskan att undvika öppna fält och söka skydd, liksom en inneboende strävan att utforska sin omgivning 6,7. Mångfalden av potentiella beteende förare bakom viljan att lösa pussel Box antyder att olika områden i hjärnan är inblandade i förmedling uppgiften prestanda. Tidigare arbete har visat att i musmodeller av schizofreni, är det prefrontala cortex samt hippocampus inblandade i förvärvet av denna uppgift 5. En lesion studie på råttor har också visat ett stort antal områden i hjärnan som är involverade i Puzzle Box prestanda, inklusive olika talamiska kärnor, hypotalamus, lillhjärnan och limbiska strukturer. Tillsammans utgör dessa fynd tyder på att engagera sig i denna problemlösning uppgiften innebär en mängd neurala strukturer i samband med kognitiv funktion.
Pusslet Box har använts framgångsrikt för att bedöma problemlösningsförmåga hos möss, samt kognitiva brister som uppvisas av murin modeller av schizofreni 5-7. Prestanda på uppgiften har visat sig vara mycket konsekvent, och korrelerar väl med resultaten från andra kognitiva beteendetester 6. Målet med detta arbete var alltså att anpassa Puzzle Box uppgiften att bli ett enkelt och tillförlitligt sätt att bestämma effektiviteten av EE.
De data som presenteras visar att Puzzle Box kan användas effektivt för att bedöma effekten av EE. Möss som tas upp i berikade miljöer konsekvent tog betydligt kortare tid att lösa obstruktion pussel inom denna beteendeanalys än gjorde djur som fötts upp inom standardlaboratorieförhållanden. Dessutom har denna skillnad var mest framträdande i den första rättegången för tre av de fyra villkor som testades, vilket tyder på EE har ett större inflytande på ett djurs infödda problemlösningsförmåga, i f?…
