Hier presenteren we een gedetailleerd protocol dat het gebruik van microvasculaire fragmenten geïsoleerd uit knaagdier- of menselijk vetweefsel schetst als een eenvoudige benadering om functioneel, gevasculariseerd beige vetweefsel te engineeren.
Engineering thermogeen vetweefsel (bijv. Beige of bruine vetweefsels) is onderzocht als een potentiële therapie voor metabole ziekten of voor het ontwerp van gepersonaliseerde microtissues voor gezondheidsscreening en medicijntests. De huidige strategieën zijn vaak vrij complex en slagen er niet in om de meercellige en functionele eigenschappen van thermogeen vetweefsel volledig weer te geven. Microvasculaire fragmenten, kleine intacte microvessels bestaande uit arteriol, venulen en haarvaten geïsoleerd uit vetweefsel, dienen als een enkele autologe bron van cellen die vascularisatie en vetweefselvorming mogelijk maken. Dit artikel beschrijft methoden voor het optimaliseren van kweekomstandigheden om het genereren van driedimensionale, gevasculariseerde en functionele thermogene vetweefsels uit microvasculaire fragmenten mogelijk te maken, inclusief protocollen voor het isoleren van microvasculaire fragmenten uit vetweefsel en kweekomstandigheden. Daarnaast worden best practices besproken, evenals technieken voor het karakteriseren van de gemanipuleerde weefsels, en monsterresultaten van zowel knaagdier als menselijke microvasculaire fragmenten worden verstrekt. Deze aanpak heeft het potentieel om te worden gebruikt voor het begrijpen en ontwikkelen van behandelingen voor obesitas en metabole ziekten.
Het doel van dit protocol is om een aanpak te beschrijven voor het ontwikkelen van gevasculariseerd beige vetweefsel uit een enkele, potentieel autologe bron, microvasculair fragment (MVF). Van bruine en beige vetweefsels is aangetoond dat ze gunstige eigenschappen vertonen die verband houden met metabole regulatie; Het kleine volume van deze vetweefseldepots bij volwassenen beperkt echter de potentiële impact op het systemische metabolisme, met name bij zieke aandoeningen zoals obesitas of diabetes type 2 1,2,3,4,5,6,7. Er is aanzienlijke belangstelling voor bruin / beige vet als een therapeutisch doelwit voor het voorkomen van de schadelijke metabole effecten in verband met obesitas en de comorbiditeiten ervan 8,9,10,11,12.
MVF’s zijn vaatstructuren die direct kunnen worden geïsoleerd uit vetweefsel, gekweekt en gedurende langere tijd in een driedimensionale configuratie kunnen worden gehouden13,14,15. Eerder werk van onze groep, en anderen, zijn begonnen met het exploiteren van de meercellige en multipotente capaciteit van MVF’s, met name als het gaat om de vorming van vetweefsel16,17,18. Als een opbouw van dit werk hebben we onlangs aangetoond dat MVF’s afgeleid van knaagdiermodellen van gezonde en type 2 diabetes19 en van menselijke proefpersonen (volwassenen ouder dan 50 jaar)20 cellen bevatten die kunnen worden geïnduceerd om thermogeen of beige vetweefsel te vormen.
Hierin is een innovatieve aanpak van waaruit een enkele bron MVF wordt gebruikt, niet alleen in staat om beige vetweefsel te creëren, maar ook de bijbehorende en kritische vasculaire component21. Het gebruik van deze techniek kan van grote waarde zijn voor studies die op zoek zijn naar een eenvoudige weefselmanipulatiebenadering voor de vorming van thermogene vetweefsels. In tegenstelling tot andere methoden die gericht zijn op het engineeren van beige vetweefsel 22,23,24,25,26,27,28, vereist het proces dat in deze studie wordt beschreven niet het gebruik van meerdere celtypen of complexe inductieregimes. Gevasculariseerde beige en witte vetmodellen kunnen worden gemaakt met MVF’s afkomstig van knaagdieren en menselijke bronnen, wat een groot vertaalpotentieel aantoont. Het eindproduct van dit protocol is een beige thermogeen vetweefsel met een structuur en metabole functie vergelijkbaar met bruin vetweefsel. Over het algemeen presenteert dit protocol het idee dat een gemakkelijk toegankelijke en mogelijk autologe bron MVF een waardevolle therapeutische interventie en hulpmiddel kan zijn voor het bestuderen van metabole stoornissen.

Obtained from Charles River (Wilmington, MA). Rats were acquired at 4 weeks of age and fed Purina 5008 until euthanasia (15-19 weeks of age). Glucose levels (blood from the lateral saphenous vein) were greater than 300 mg/dL in all FA/FA rats used in the study.
