Genaamd Een techniek<strong> C</strong> Omprehensive<strong> M</strong> Icroarray<strong> P</strong> Olymer Profiling (CoMPP) voor de karakterisering van plantencelwand glycanen wordt beschreven. Deze methode combineert de specificiteit van monoklonale antilichamen gericht tegen bepaalde epitopen-glycan met een miniatuur microarray analyse platform zodat screening van glycan voorkomen in een breed scala aan biologische context.
Plant celwanden zijn complexe matrices van heterogene glycanen die een belangrijke rol in de fysiologie en ontwikkeling van planten te spelen en bieden de grondstoffen voor de menselijke samenleving (bv hout, papier, textiel en biobrandstof-industrie) 1,2. Echter, het begrijpen van de biosynthese en functie van deze componenten blijft uitdagend.
Celwand glycanen zijn chemisch en conformationeel gevarieerd vanwege de complexiteit van hun bouwstenen, de glycosyl residuen. Deze vormen bindingen op meerdere posities en verschillen in ringstructuur isomere of anomere configuratie, en bovendien zijn gesubstitueerd met een array van niet-suikerresten. Glycaan samenstelling varieert in verschillende cel-en / of weefseltypen of subgebieden van een celwand 3. Bovendien is de samenstelling ook aangepast tijdens de ontwikkeling 1 of in reactie op omgevingsfactoren 4.
In exproces van 2.000 genen Plant celwanden zijn complex arrays heterogene glycanen voorspeld betrokken bij celwand glycan biosynthese en modificatie in Arabidopsis 5. Echter, relatief weinig biosynthetische genen functioneel 4,5 gekarakteriseerd. Reverse genetics benaderingen moeilijk omdat de genen vaak differentieel tot expressie, vaak op lage niveaus tussen celtypes 6. Ook zijn mutant studies vaak belemmerd door gen redundantie of compenserende mechanismen om te zorgen voor passende celwand functie is 7 gehandhaafd. Zo nieuwe benaderingen nodig zijn om snel te karakteriseren van de uiteenlopende glycaanstructuren en functionele genomica gebaseerde benaderingen bevorderen om te begrijpen celwand biosynthese en modificatie.
Monoklonale antilichamen (mAbs) 8,9 naar voren zijn gekomen als een belangrijk instrument voor het bepalen van glycan structuur en de verdeling in planten. Deze herkent distInCT epitopen aanwezig in grote klassen van plantencelwand glycanen, met inbegrip van pectines, xyloglucanen, xylanen, mannanen, glucanen en arabinogalactans. Onlangs het gebruik uitgebreid tot grootschalige screening experimenten om de relatieve abundantie van glycanen bepalen in een breed scala van planten en weefseltypes tegelijkertijd 9,10,11.
Hier wordt een microarray gebaseerde glycan screening methode genaamd Comprehensive Polymer Microarray Profiling (CoMPP) (figuren 1 en 2) 10,11 die meerdere monsters kan (100 sec) worden gescreend met een geminiaturiseerde microarray platform met beperkte reagens en monstervolumes. De plek signalen op de microarray kan formeel worden gekwantificeerd aan semi-kwantitatieve gegevens over glycan epitoop optreden te geven. Deze benadering is geschikt voor het bijhouden glycan veranderingen in complexe biologische systemen 12 en een globaal overzicht van celwandsamenstelling bijzonder wanneer voorkennis of deze niet beschikbaar is.
CoMPP is een snelle en gevoelige werkwijze voor het profileren van de glycan samenstelling van honderden plantaardige monsters in een paar dagen. Deze methode aanvulling op de reeds beschikbare bacteriën of zoogdiercellen glycan matrix platforms voor high-throughput screening van koolhydraat interacties met glycan-binding zoals lectines, receptors en antilichamen 16.
