The goal of this protocol is to use laser-capture micro-dissection as an effective method to isolate pure populations of cell types from heterogeneous prostate tissues for downstream RNA analysis.
The prostate gland contains a heterogeneous milieu of stromal, epithelial, neuroendocrine and immune cell types. Healthy prostate is comprised of fibromuscular stroma surrounding discrete epithelial-lined secretory lumens and a very small population of immune and neuroendocrine cells. In contrast, areas of prostate cancer have increased dysplastic luminal epithelium with greatly reduced or absent stromal population. Given the profound difference between stromal and epithelial cell types, it is imperative to separate the cell types for any type of downstream molecular analysis. Despite this knowledge, the bulk of gene expression studies compare benign prostate to cancer without micro-dissection, leading to stromal bias in the benign samples. Laser-capture micro-dissection (LCM) is an effective method to physically separate different cell types from a specimen section. The goal of this protocol is to show that RNA can be successfully isolated from LCM-collected human prostatic epithelium and used for downstream gene expression studies such as RT-qPCR and RNAseq.
Prostata er en heterogen væv sammensat af sekretorisk epitel anbragt i glandulær acini omgivet af fibromuskulær stroma består primært af glat muskulatur 1. Epitel rum består af fem forskellige, men organiserede celletyper: basale celler, sekretoriske celler, neuroendokrine celler, transit forstærkende celler og stamceller 2. I prostatakræft (PCA), der opstår fra luminale epitelceller, væksten af adenocarcinom forårsager en tydelig gradvist tab af stromale rum 3. Af disse grunde vil vævsprøver have tydelige forskelle i andelen af stromale og epitelial celletype baseret på omfanget af PSA. Disse forskelle kan føre til tendentiøse antagelser af genekspression data indhentet fra hele væv uden hensyn til mikrodissektion af den ønskede celletype. Derfor, for at fjerne denne skævhed er det vigtigt at adskille celletyper før RNA-ekstraktion oggenekspression analyse.
Macrodissection eller mikrodissektion kan anvendes til fysisk adskilt velkarakteriserede epitel områder fra det omgivende stroma 4 -6. Macrodissection sker typisk med et barberblad under et dissektionsmikroskop og fungerer godt til adskillelse af store PCa knuder fra stroma, men ikke er i stand til at fjerne godartede epitel fra omgivende stroma (se eksempel på benign prostata histologi i figur 1). Mikrodissektion med en laser (LCM) er betydeligt mere arbejdskrævende end macrodissection, men kan meget præcist dissekere godartet epitel 4.
Nylige publikationer fra vores laboratorium har vist, at RNA kan med held udvindes af LCM fra enten formalinfikserede paraffinindlejrede (FFPE) biopsier eller frosne væv 4,7-9. De store udfordringer i LCM-RNA-ekstraktion er 1) til nøjagtigt at dissekere de ønskede områder af vævet, og 2) at bevare RNA integritet under LCM og isolation proces 4,10. RNA isoleret fra rene cellepopulationer kan anvendes til genekspression analyse af flere metoder, herunder revers-transkription kvantitativ PCR (RT-qPCR) 7,8, microarray 11, og dyb -sequencing 12-14.
Målet med denne protokol er at isolere total RNA fra LCM prostata epitel fra frosne væv til nedstrøms genekspression analyser.
Genekspression profilering fra humane prøver kan være udfordrende, ikke kun for kvaliteten eller kvantiteten af væv til rådighed, men også for de forskellige histologiske enheder til stede i en given vævsprøve. Dette er en særlig udfordring i prostata, hvor godartede væv er stort set stromale væv og områder af kræft er blottet for stroma.
