Abstract
Utendørs aerosol forskning bruker ofte svevestøv samples på filtre. Denne fremgangsmåten gjør det mulig for forskjellige karakteristikk av de oppsamlede partikler som skal utføres i parallell. Hensikten med fremgangsmåten som presenteres her, er å oppnå en meget nøyaktig og pålitelig analyse av endotoksin og DNA-innhold av bio-aerosoler ekstrahert fra filtrene. Utvinningen av høy molekylvekt, organiske molekyler, slik som lipopolysakkarider, fra samplede filtre innbefatter risting av prøven i et pyrogenfritt vann-basert medium. Den etterfølgende analyse bygger på en enzymatisk reaksjon som kan detekteres ved hjelp av en turbidimetrisk måling. Introduction
Luft sampling på filtrene er et grunnleggende verktøy i atmosfære aerosoler forskning. 1 De samplede filtrene er utgangspunktet for en rekke kjemiske, fysiske og biologiske karakterisering av de innsamlede omgivelses partiklene. 2-11 Fordelen med denne tilnærmingen er at ulike analyser kan bli utføres off-line på samme prøve. Sammenstille dataene fra alle de ulike analyser gjør det mulig for forskeren å få en god forståelse av hva som kjennetegner de innsamlede partikler og hjelpemidler i å løse komplekse spørsmål i atmosfærisk vitenskap. 12,13 for eksempel marine og innland luftprøver tatt i løpet av samme periode kan sammenlignes med hensyn til det samplede partikkel giftighet og biologisk preparat. 14 for denne studien, lipopolysakkarider (LPS) komponenter på gram-negative bakteriecellevegger, også kjent som endotoksiner, ble ekstrahert fra filtrene samplet på land og ved en innlandet området, og ble evaluert ved bruk avLimulus amebocyttlysat (LAL) test. Parallelt ble en genomisk evaluering av bakterieinnhold (totalt antall bakterier, gram-negative, og cyanobakterier) ble utført på den samme prøven ved hjelp av kvantitativ polymerasekjedereaksjon (q-PCR). LAL-testen er basert på målinger av turbiditet som dannes etter tilsetning av et vandig ekstrakt av amebocytes fra hesteskokrabbe, Limulus Polyfemos, til en vandig oppløsning som inneholder endotoksiner. Jo høyere endotoksin-konsentrasjonen i prøven, utvikler raskere turbiditet. 15 q-PCR-analyse er basert på en fluorescens-signal som sendes ut som et spesifikt DNA-fragment blir amplifisert. 16 Av sanntids overvåkning av signalet i løpet av den eksponensielle fase av PCR reaksjon og kalibrerings med en standardkurve, kan den opprinnelige mengde DNA kvantifiseres.
