Abstract
Gleichzeitige Sauerstoffversorgung und Überwachung der Glucosestimulus-Sekretion Kopplungsfaktoren in einer einzigen Technik ist entscheidend für die Modellierung von pathophysiologischen Zuständen von Insel Hypoxie, vor allem in der Transplantations-Umgebungen. Standard-hypoxischen Kammer Techniken können nicht beide Reize modulieren gleichzeitig noch eine Echtzeit-Überwachung des Glucosestimulus-Sekretion Kopplungsfaktoren. Um diese Schwierigkeiten zu begegnen, haben wir mit einer mehrschichtigen Mikrofluidtechnik, sowohl wässrige und Gasphasenmodulationen über eine Diffusionsmembran integrieren. Dies erzeugt einen Stimulations Sandwich um die mikroskalierte Inseln innerhalb des transparenten Polydimethylsiloxan (PDMS)-Vorrichtung, um so die Überwachung der genannten Kopplungsfaktoren über Fluoreszenzmikroskopie. Zusätzlich wird die Gaszufuhr durch ein Paar von Mikrodispenser gesteuert wird, eine quantitative, Unter Minute Modulationen von Sauerstoff zwischen 0-21%. Diese intermittierende Hypoxie wird angewendet, um ein neues Phänomen der Insel zu untersuchent Präkonditionierung. Außerdem mit multimodalen Mikroskopie bewaffnet, waren wir in der Lage, auf detaillierte Kalzium und K ATP-Kanal Dynamik in diesen hypoxischen Ereignissen zu suchen. Wir haben die Vision mikrofluidischen Hypoxie, vor allem diese simultane Dualphasen Technik, die als ein wertvolles Instrument bei der Untersuchung Inseln sowie viele Ex-vivo-Gewebe.
